摘　　要：目的探究肺耐药蛋白(LRP)、Twist蛋白表达与结直肠癌化疗耐药及预后的关系。方法体外培养结直肠癌SW480,使用siRNA敲低LRP和Twist的表达;并分为正常对照组、干扰LRP组和干扰Twist组;使用蛋白质印记法分别检测各组细胞LRP蛋白和Twist蛋白表达情况;使用MTT检测各组细胞对化疗药物的敏感性,并检测其增殖蛋白Ki67表达情况;使用Transwell检测各组细胞侵袭能力;收集2012年1月至2014年1月于武警宁夏总队医院治疗的结直肠癌患者37例,取其肿瘤样本和癌旁组织,使用蛋白质印记法检测癌组织和癌旁组织LRP蛋白及Twist蛋白表达情况;将患者根据LRP蛋白和Twist蛋白表达情况分为LRP阳性组(25例)、LRP阴性组(12例)和Twist蛋白阳性组(22例)、Twist蛋白阴性组(15例),分别统计各组患者3年生存率。结果相比正常对照组,干扰LRP组LRP蛋白相对表达水平、Ki67蛋白相对表达水平、单位面积侵袭细胞数目显著降低(P<0.05),Twist蛋白相对表达无显著差异(P>0.05);相比正常对照组,干扰Twist组Twist蛋白相对表达水平、Ki67蛋白相对表达水平、单位面积侵袭细胞数目显著降低(P<0.05),LRP蛋白相对表达无显著差异(P>0.05);MTT实验发现干扰LRP组和干扰Twist组细胞耐药性显著降低(P<0.05);LRP阳性组3年生存率为60.00%显著低于LRP阴性组的91.60%(2=3.892,P=0.049);Twist蛋白阳性组3年生存率为54.55%显著低于Twist蛋白阴性组的93.33%(2=6.423,P=0.011)。结论结直肠癌患者癌组织LRP蛋白和Twist蛋白相对表达水平升高;降低LRP蛋白和Twist蛋白表达可以有效抑制直肠癌癌细胞的耐药性,延长患者生存期。Objective To explore relationship between expression of lung resistance protein(LRP),Twist protein and drug resistance,prognosis of colorectal cancer chemotherapy.Methods Colorectal cancer cells SW480 were cultured in vitro.The expressions of LRP and Twist was knocked down by siRNA.They were divided into normal control group,interference LRP group and interference Twist group.Western blot was applied to detect expression of LRP and Twist protein in each group.The sensitivities of cells to chemotherapy drugs in each group were detected by MTT.The expression of proliferin Ki67 was detected.The cells invasion ability in each group was detected by Transwell.Thirty-seven patients with colorectal cancer who were admitted to the hospital from January 2012 to January 2014 were enrolled.The tumor samples and para-carcinoma tissues were collected.Western blot was applied to detect expression of LRP and Twist protein in in cancerous tissues and para-carcinoma tissues.According different expression of LRP and Twist protein,patients were divided into positive LRP group(25 cases),negative LRP group(12 cases),positive Twist protein group(22 cases)and negative Twist protein group(15 cases).The 3-year survival rates in all groups were statistically analyzed.Results Compared with normal control group,relative expression levels of LRP and Ki67 protein,and number of invaded cells per unit area were significantly increased in interference LRP group(P<0.05).The difference in relative expression of Twist protein was not statistically significant between the two groups(P>0.05).Compared with normal control group,relative expression levels of Twist and Ki67 proteins,and number of invaded cells per unit area were significantly decreased in interference Twist group(P<0.05).The difference in relative expression of LRP was not statistically significant between the two groups(P>0.05).MTT experiment found that cell drug resistance was significantly decreased in interference LRP group and interference Twist group(P<0.05).The 3-year survival rate
