摘  要:目的观察紫河车提取物对人脑胶质瘤细胞株U251增殖、侵袭和迁移的影响,及其对粘着斑激酶(focal adhesion kinase,FAK)表达水平的影响。方法把正常培养传代后的U251胶质瘤细胞随机为对照组、紫河车组(给予人胎盘组织液400mg/mL)。采用MTT法和软琼脂克隆形成实验分析细胞增殖能力,Transwell小室检测细胞侵袭能力,划痕实验检测细胞迁移能力。结果细胞培养48h和72h时,紫河车组U251细胞增殖能力低于对照组(P<0.05)。紫河车组U251细胞侵袭能力和迁移能力均低于对照组,差异有统计学意义(P<0.05)。紫河车组U251细胞FAK和p-FAK mRNA和蛋白表达量低于对照组,差异有统计学意义(P<0.05)。结论紫河车提取物可能通过FAK信号通路抑制脑胶质瘤细胞的增殖、侵袭和迁移。Objective To observe the effect of extract of Ziheche on proliferation,invasion and migration of human glioma cell line U251 and the expression of focal adhesion kinase(FAK).Methods U251 glioma cells were randomly divided into control group and Ziheche group(400 mg/mL of human placental tissue fluid).MTT assay and soft agar clone formation assay were used to analyze cell proliferation,Transwell chamber was used to detect cell invasion,and scratch test was used to detect cell migration.Results The proliferation of U251 cells in Ziheche group was lower than that in control group at 48h and 72h(P<0.05).The invasive ability and migratory ability of U251 cells in Ziheche group were lower than those in the control group(P<0.05).The expression of FAK and p-FAK in U251 cells of Ziheche group was lower than that of the control group(P<0.05).Conclusion The extract of Ziheche may inhibit the proliferation,invasion and migration of glioma cells through FAK signaling pathway.
