摘 要:为优化虎杖根大黄素的最佳提取工艺条件,采用三因素三水平的Box-Behnken试验设计,通过Design-Expert软件在单因素试验的基础上,以提取温度、乙醇体积分数和提取时间为自变量,大黄素的提取率为响应值,通过回归分析各工艺参数与响应值之间的关系,由此预测虎杖根大黄素最佳提取工艺。结果表明:最佳提取工艺条件为乙醇体积分数75.5%、提取时间30 min、提取温度40.6℃、料液比1︰20 (g/mL),虎杖根大黄素的提取率为0.577%,与预测值的相对误差为0.35%,采用响应面法优化虎杖中大黄素提取工艺条件是合理可行的。The response surface method was mainly by three factors and three levels of Box-Behnken experimental design. Through the Design-Expert software on the basis of single factor experiment, ethanol concentration, extraction temperature, and extraction time were used by three factors as independent variables, the extraction yield of emodin from Polygonum cuspidatum was used as the response value, through the regression analysis of the relationship between the various process parameters and the response value, in order to predict the best process conditions. The result showed that the optimum extraction conditions of emodin from Polygonum cuspidatum were as follows as: 75.5% of ethanol concentration, 30 min of extraction time, 40.6 ℃ of extraction temperature, and 1︰20(g/mL) of material liquid. Under the optimal conditions, the extraction rate of emodin from Polygonum cuspidatum was 0.577%, and the relative error between predictive value and experimental value was 0.35%. The experiments showed that the response surface method optimized by extraction process of emodin in the giant knotweed condition was feasible.
