磷酸二酯酶4各亚型mRNA在特应性皮炎及银屑病皮损中的表达水平及其在发病机制中的作用

摘　　要：目的探讨磷酸二酯酶4(PDE4)各亚型mRNA在特应性皮炎(AD)及银屑病皮损组织中的表达水平及其在发病机制中的作用。方法选取21例AD患者(AD组)、19例银屑病患者(银屑病组)及15例皮肤健康正常者(对照组)作为研究对象。采用实时荧光定量PCR反应法检测PDE4各亚型(PDE4A、PDE4B、PDE4C、PDE4D)mRNA的相对表达水平。结果 AD组患者皮损中PDE4B mRNA、PDE4D mRNA相对表达水平高于对照组(P<0.05);银屑病组患者皮损组织中PDE4A mRNA、PDE4D mRNA相对表达水平高于对照组(P<0.05);AD组患者皮损组织中PDE4B mRNA相对表达水平呈特异性高表达(P<0.05),银屑病组患者皮损组织中PDE4A mRNA相对表达水平呈特异性高表达(P<0.05)。结论 PDE4各亚型mRNA在AD及银屑病皮损组织中的表达水平呈现差异性,这可能在AD和银屑病的发生及病情演变过程中发挥重要作用。Objective To explore the expression levels of phosphodiesterase 4(PDE4) subtypes mRNAs in skin lesion tissue in atopic dermatitis(AD) and psoriasis as well as their role in the pathogenesis. Methods Twenty-one AD patients(AD group), 19 psoriasis patients(psoriasis group) and 15 individuals with healthy skin(control group) were selected as research subjects. Real-time fluorescence quantitative PCR assay was used to detect the relative expression levels of PDE4 subtypes(PDE4 A, PDE4 B, PDE4 C,and PDE4 D) mRNAs. Results The relative expression levels of PDE4 B and PDE4 D mRNAs in skin lesions of the AD group were higher than those of the control group(P<0.05);patients in the psoriasis group exhibited higher relative expression levels of PDE4 A and PDE4 D mRNAs in skin lesion tissue than the control group(P<0.05);the relative expression level of PDE4 B mRNA presented a high-expression specificity in skin lesion tissue of patients in the AD group(P<0.05),and the relative expression level of PDE4 A mRNA exhibited a high-expression specificity in skin lesion tissue of patients in the psoriasis group(P<0.05). Conclusion The expression levels of PDE4 subtypes mRNAs demonstrate difference between AD and psoriasis skin lesion tissue, which may play an important role in the occurrence and evolution of AD and psoriasis.
