摘 要:目的探究姜黄素对子宫内膜癌HEC-1-B细胞增殖、侵袭能力及相关蛋白的影响。方法低、中、高剂量实验组及对照组子宫内膜癌HEC-1-B细胞分别以30、60、120μmol·L^-1姜黄素处理,各组分别处理24、36、48 h,噻唑蓝(MTT)法检测HEC-1-B细胞增殖抑制情况;Transwell小室实验检测HEC-1-B细胞侵袭能力;蛋白免疫印迹(WB)法检测HEC-1-B细胞基质金属蛋白酶-2、9(MMP-2、MMP-9)及细胞周期蛋白D1(Cyclin D1)蛋白表达情况。结果低、中、高剂量实验组HEC-1-B细胞增殖抑制率均随着姜黄素作用浓度增大及作用时间延长呈逐渐升高趋势,侵袭细胞数量、侵袭率及MMP-2、MMP-9及Cyclin D1蛋白相对表达量均随姜黄素作用浓度增大呈逐渐降低趋势,且均低于对照组,两两之间比较差异均有统计学意义(P均<0.05)。结论姜黄素可有效抑制子宫内膜癌HEC-1-B细胞增殖,降低其侵袭能力,同时有效抑制MMP-2、MMP-9及Cyclin D1蛋白的表达,发挥抗子宫内膜癌作用。Objective To investigate the effects of curcumin on the proliferation,invasion of endometrial cancer HEC-1-B cells and expression of related proteins. Methods Endometrial cancer HEC-1-B cells in experimental-L/M/H groups and control group were treated with the final concentration of 30,60,120 μmol·L^-1 curcuma and equal amount of normal saline,each group was intervened for 24,36 and 48 hours. The proliferation inhibition,the invasion ability and the expression of matrix metalloproteinases 2,9(MMP-2,MMP-9)and cell cycle protein D1(Cyclin D1)in HEC-1-B cells were detected by methyl thiazolyl tetrazolium(MTT),transwell chamber test and Western blot(WB)method,respectively. Results The cell proliferation inhibition rates of HEC-1-B cells in experimental-L/M/H groups increased gradually with the increase of curcuma action concentration and the extended of action time,the invasive cell number,invasive rate and the levels of MMP-2,MMP-9,Cyclin D1 protein relative expression decreased with the increase of curcumin concentration in experimental-L/M/H groups,and which were lower than those in control group,with significant differences among each group(P all<0.05). Conclusion Curcumin can inhibit the endometrial cancer HEC-1-B cells proliferation effectively,and reduce the invasive ability,and it can effectively inhibit the expression of related proteins MMP-2,MMP-9 and Cyclin D1,plays an anti-endometrial cancer effect.
