摘  要:目的探讨丙酮酸钠对严重烫伤多器官功能障碍综合征(MODS)小鼠胰岛β细胞功能不全的保护作用。方法选用昆明小鼠140只,制作30%体表面积Ⅲ度严重烫伤MODS小鼠模型,随机分为健康组20只,丙酮酸钠治疗组和生理盐水对照组各60只。3组小鼠在伤后1、3、5 d心脏采血检测丙氨酸转氨酶(ALT)、血肌酐(Cr)、肌酸激酶MB型同工酶(CK-MB)、白介素-6(IL-6)、超氧化物歧化酶(SOD)、血糖和稳态模型β细胞功能指数(HOMA-β)水平。结果丙酮酸钠治疗组伤后1、3、5 d血清ALT、Cr、CK-MB、IL-6水平均低于生理盐水对照组,HOMA-β高于生理盐水对照组(P<0.01);丙酮酸钠治疗组伤后3、5 d血SOD水平均高于生理盐水对照组(P<0.01)。结论严重烫伤后MODS小鼠会出现高血糖及炎性反应,丙酮酸钠可能有助于对胰腺组织的保护作用,改善小鼠胰岛β细胞功能。Objective To investigate the effect of sodium pyruvate on isletβ-cell dysfunction in the mice with multiple organ dysfunction syndrome(MODS)induced by severe scald.Methods A total of 140 Kunming mice were selected to make MODS mouse models with 30%total body surface areaⅢdegree scalds.They were randomly divided into healthy group(n=20),sodium pyruvate treatment group(n=60)and normal saline control group(n=60).Blood samples were collected from the mice in the three groups at 1,3,and 5 days after injury to detect alanine aminotransferase(ALT),blood creatinine(Cr),creatine kinase-MB(CK-MB),interleukin-6(IL-6),superoxide dismutase(SOD),glucose and homeostasis model assessmentβ-cell function(HOMA-β)levels.Results The levels of serum ALT,Cr,CK-MB and IL-6 in the sodium pyruvate treatment group were lower than those in the saline control group at 1,3 and 5 days after injury,and HOMA-βwas higher than that in the saline control group(P<0.01)The sodium SOD level in the sodium pyruvate treatment group was higher than that in the normal saline control group at 3 and 5 days after injury(P<0.01).Conclusion Hyperglycemia and inflammatory response may occur in MODS mice after severe scald.Sodium pyruvate may help protect pancreatic tissues and improve the function of isletβcells in mice.
