不同病理类型食管癌GRP78、GRP94表达及与预后的关系分析

摘　　要：目的分析不同病理类型食管癌葡萄糖调节蛋白78(GRP78)、94(GRP94)表达及与预后的关系。方法收集2010年1月-2014年1月手术治疗的78例食管癌石蜡包埋癌组织和30例癌旁石蜡包埋组织,观察不同病理类型食管癌GRP78、GRP94表达情况,检测不同病理类型食管癌组织中GRP78、GRP94 mRNA和蛋白相对表达水平,计算随访患者术后生存率。结果食管癌腺癌组织GRP78和GRP94高表达率均高于食管鳞癌组织和癌旁组织,且鳞癌组织高于癌旁组织(P<0.05)。食管癌腺癌组织GRP78 mRNA、GRP78蛋白相对表达量明显高于食管癌鳞癌组织(P<0.01)。GRP78高表达率与临床分期、分化程度、病理类型有关,而GRP94高表达率与临床分期、分化程度有关(P<0.05)。GRP78和GRP94低表达组术后3年生存率高于GRP78和GRP94高表达组(P<0.05)。结论不同病理类型食管癌组织中GRP78、GRP94表达水平存在明显差异,食管癌患者术后生存率与GRP78、GRP94表达水平密切相关,GRP78、GRP94或可作为食管癌病理类型及预后评估的有效指标。Objective To analyze expression of glucose regulatory protein 78(GRP78)and 94(GRP94)in different pathological types of esophageal cancer(EC)and its relationship with prognosis.Methods The paraffin-embedded tissues from 78 patients with EC and adjacent paraffin-embedded tissues from 30 patients who underwent surgery in the hospital from January 2010 to January 2014 were collected.The expression of GRP78 and GRP94 in different pathological types of EC was observed.The relative expression of GRP78,GRP94 mRNA and protein in different pathological types of EC was detected.The postoperative survival rate of patients was calculated.Results The high expression rates of GRP78 and GRP94 in esophageal adenocarcinoma tissues were higher than those in esophageal squamous cell carcinoma and adjacent tissues,and the rates in squamous cell carcinoma tissues were higher than those in adjacent tissues(P<0.05).The relative expression levels of GRP78 mRNA and GRP78 protein in esophageal adenocarcinoma tissues were significantly higher than those in esophageal squamous carcinoma tissues(P<0.01).The high expression rate of GRP78 is related to clinical stage,degree of differentiation and pathological type,while the high expression rate of GRP94 is related to clinical stage and degree of differentiation(P<0.05).The 3-year survival rate of the GRP78 and GRP94 low expression group was higher than that of the GRP78 and GRP94 high expression group(P<0.05).Conclusion There are significant differences in the expression levels of GRP78 and GRP94 in different pathological types of EC.The postoperative survival rate of patients with EC is closely related to expression levels of GRP78 and GRP94.GRP78 and GRP94 can be used as effective indexes for evaluating the pathological types and prognosis of EC.
