摘 要:目的研究柚皮苷是否通过调控ARHI基因表达影响结肠癌细胞的增殖和凋亡。方法采用不同浓度的柚皮苷处理结肠癌细胞SW620,MTT法检测细胞增殖,免疫印迹实验(Western blotting)检测细胞周期蛋白D1(CyclinD1)、p21、B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、ARHI蛋白表达,流式细胞术检测细胞凋亡,实时定量PCR(qRT-PCR)检测ARHI mRNA表达。在SW620细胞中转染pcDNA-ARHI,或转染si-ARHI,并使用柚皮苷处理,观察其对细胞增殖和凋亡的影响。结果与对照组比较,柚皮苷明显增加SW620细胞的增殖抑制率、凋亡率、p21、Bax蛋白表达量、ARHI mRNA和ARHI蛋白表达量,显著降低CyclinD1、Bcl-2蛋白水平,均呈浓度依赖性(P<0.05)。ARHI过表达明显增加SW620细胞的抑制率、凋亡率、p21、Bax蛋白表达量,显著降低CyclinD1和Bcl-2蛋白水平(P<0.05)。抑制ARHI表达逆转了柚皮苷对SW620细胞增殖抑制率、凋亡率、p21、Bax蛋白表达的促进作用,及对CyclinD1和Bcl-2蛋白表达的抑制作用。结论柚皮苷通过上调ARHI基因的表达,抑制结肠癌细胞增殖,并诱导细胞凋亡。Objective To investigate whether Naringin affects proliferation and apoptosis of colon cancer cell by regulating the expression of ARHI gene. Methods Colon cancer cells SW620 were treated with different concentrations of Naringin, and MTT was used to detect cell proliferation. Western blotting was employed to analyze the expressions of CyclinD1, p21, Bcl-2, Bax and ARHI. Cell apoptosis was determined by flow cytometry. qRT-PCR was used to measure the expression of ARHI mRNA. SW620 cells were transfected with pcDNA-ARHI, or transfected with si-ARHI and treated with Naringin to observe the effects on cell proliferation and apoptosis. Results Compared with control group, Naringin obviously increased the proliferation inhibition rate, apoptosis rate, expressions of p21, Bax protein, ARHI mRNA, and ARHI protein in SW620 cells, and significantly reduced the levels of CyclinD1 and Bcl-2 proteins, all were concentration-dependent(P<0.05). ARHI overexpression remarkably increased the inhibition rate, apoptosis rate, expressions of p21, and Bax protein of SW620 cells, and dramatically reduced levels of CyclinD1 and Bcl-2 proteins(P<0.05). Inhibition of ARHI expression reversed the effect of Naringin on the proliferation inhibition rate, apoptosis rate, expressions of p21, Bax proteins of SW620 cells, and the inhibitory effect of CyclinD1 and Bcl-2 proteins.Conclusion Naringin inhibits proliferation of colon cancer cell and induces apoptosis by up-regulating the expression of ARHI gene.
