摘  要:以挥发油提取率为评价指标,采用单因素实验和响应面法实验优化陕产长春七挥发油的超声提取工艺,并考察了长春七挥发油对DPPH自由基和ABTS自由基的清除能力,评价其抗氧化活性。确定长春七挥发油的最佳提取工艺为:以乙醚为提取溶剂、料液比1∶53(g∶mL)、超声时间20 min、超声温度26℃,在此工艺条件下,挥发油提取率为13.6392%,与理论值相差0.0024%,说明此方法可行。长春七挥发油对DPPH自由基和ABTS自由基均有较高的清除率,且随着浓度增大清除能力增强,长春七挥发油清除DPPH自由基线性范围在1.4~1.8 mg·mL^-1为最佳,长春七挥发油清除ABTS自由基线性范围在2.5~3.5 mg·mL^-1为最佳。为进一步有效开发利用陕西“太白七药”提供了一定的理论基础。Using the extraction rate of volatile oil as an evaluation index,we optimized the ultrasonic extraction process of volatile oil from Libanotis buchtorimensis of Shaanxi by single-factor experiments and response surface methodologies.Furthermore,we investigated the scavenging abilities of volatile oil from Libanotis buchtorimensis on DPPH free radicals and ABTS free radicals,and evaluated their antioxidant activities.The optimum extraction process of volatile oil from Libanotis buchtorimensis is determined as follows:using ethyl ether as the extraction solvent,the solid-liquid ratio of 1∶53(g∶mL),the ultrasonic time of 20 min,and the ultrasonic temperature of 26℃.Under above conditions,the actual extraction rate is 13.6392%,which differs by 0.0024%from the theoretical value,indicating that this method is feasible.The volatile oil from Libanotis buchtorimensis has higher scavenging rates of DPPH free radicals and ABTS free radicals,and the scavenging ability increases with the increase of concentration.The volatile oil from Libanotis buchtorimensis has the best DPPH scavenging ability in the linear range of 1.4-1.8 mg·mL^-1,while it has the best ABTS scavenging ability in the linear range of 2.5-3.5 mg·mL^-1.This study provides a theoretical basis for the further effective development and utilization of Shaanxi"Taibai Qiyao".
