摘　　要：他拉唑帕尼于2018年10月16日通过FDA加速审批程序批准上市,是FDA批准上市的第4个聚腺苷二磷酸核糖聚合酶[poly(ADP-ribose) polymerase,PARP]抑制剂,用于治疗有害或疑似有害生殖系BRCA突变HER2阴性局部晚期或转移性乳腺癌的成人患者。本文对他拉唑帕尼的药理作用、药动学、毒理学、临床研究、安全性等进行了归纳总结。Talazoparib,approved under accelerated approval by the FDA on Oct 16,2018,is the fourth poly(ADP-ribose)polymerase(PARP)inhibitor approved by FDA.It is indicated for the treatment of adult patients with deleterious or suspected deleterious germline breast cancer susceptibility gene-mutated(BRCA)human epidermal growth factor receptor 2(HER2)-negative locally advanced or metastatic breast cancer.This review summarizes the pharmacology,pharmacokinetics,nonclinical toxicity,clinical trials,and safety of talazoparib.
