四角赖利绦虫(Raillietina tetragona)ITS-1 rDNA序列的扩增及分析

摘　　要：鸡赖利氏绦虫病是由四角赖利氏绦虫(Raillietina tetragona)、棘沟赖利氏绦虫(Raillietina echinobothrida)和有轮赖利氏绦虫(Raillietina cesticillus)寄生于鸡的小肠所引起的一类疾病。该病呈全球性分布,严重阻碍了养禽业的发展。本研究通过PCR扩增来自湖南省的10条四角赖利氏绦虫的核糖体DNA第一内转录间隔区(first internal transcribed spacers,ITS-1) ITS-1 rDNA,阳性产物经纯化、测序,最后获得1 268 bp的ITS-1序列,对所获得的序列进行分析。结果表明:四角赖利氏绦虫ITS-1的种内差异为0%~0.1%,种间差异为25. 22%~26.48%,说明ITS-1相对保守,可作为研究种间遗传变异的标记。Chicken cestodosis is a disease caused by Raillietina tetragona,Raillietina echinobothrida and Raillietina cesticillus parasitizinginsmall intestine of chicken.This disease is wide distribution in all the around of world,and has seriously hindered the development of poultry industry.In the present study,the first internal transcribed spacers(ITS-1)of 10 strains of Raillietina tetragona from Hunan province were amplified by polymerase chain reaction(PCR).All positive PCR products were purified and sequenced,and the sequences of ITS-1 region have a size of 1268 bp.All the sequences were analyzed.The results showed that the intra-specific variations within Raillietina tetragona ranged from 0%to 0.1%,and the intre-specific variations from 25.22%to 26.48%,which suggested that the ITS-1 sequence was conserved and might be used as an useful genetic marker for population genetics studies.The results of this study will provide reference and basis for further studying molecular identification,population genetics of Raillietina tetragona.
