摘 要:探讨促红细胞生成素(EPO)在增殖性视网膜新生血管形成中的作用。采用高氧诱导的方法制作小鼠增殖性视网膜病变模型,运用视网膜铺片免疫组化染色观察视网膜新生血管的形态。用RT-PCR及Western blot技术测定视网膜组织中EPO的表达水平。结果显示,模型鼠视网膜组织具有大片无灌注区和视网膜新生血管形成伴荧光渗漏。模型鼠视网膜组织中EPO mRNA及蛋白质表达水平在缺氧状态下逐渐升高,于缺氧48小时(出生后14天)到达高峰,然后缓慢下降,至出生后26天降至基线水平。这说明,氧诱导的增殖性视网膜病变模型鼠中,持续缺氧的视网膜组织通过增加EPO的表达,从而诱导视网膜新生血管的发生。To explore the effect of erythropoietin(EPO)on the development of retinal neovascularization in the proliferative retinopathy.The oxygen-induced proliferative retinopathy model was set up by hyperoxia exposure.Retinal flatmount followed by immunohistochemistry was performed to observe retinal neovascularization.The expression of EPOin the murine retina was detected by reverse transcription polymerase chain reaction(RT-PCR)and Western blot.The results showed,murine retina of model group displayed large non-perfusion area and retinal neovascularization with fluoresceince leakage,the expression of EPO mRNA and protein increased after retinal hypoxia occurred,it reached the peak after 48 hours hypoxia exposure,and then decreased to baseline on postnatal day 26.In the murine oxygen-induced proliferative retinopathy model,sustained hypoxia retinal cells promoted production of EPO,which facilitated pathological retinal angiogenesis.
