FOXC1和FOXCUT在肝细胞癌组织中的表达及与预后的关系

摘　　要：目的:探讨肝细胞癌组织中FOXC1基因以及上游启动子非编码蛋白FOXCUT的表达情况,并进行相关性分析。方法:选取2013年9月至2015年6月我院收治的肝细胞癌患者62例作为研究对象,取其手术过程中切除的癌组织及癌旁组织,采用qPCR法检测FOXC1 mRNA、lncRNA FOXCUT在肝细胞癌中的表达,采用免疫组化法测定临床石蜡切片中FOXC1蛋白的表达,分析FOXC1、FOXCUT与临床病理特征的关系,Pearson法分析FOXC1与FOXCUT之间相关性,Kaplan-Meier法分析肝细胞癌组织中FOXC1、FOXCUT表达与患者生存率的关系。结果:两组FOXC1、FOXCUT在肝细胞癌中表达水平存在明显差异,与癌旁组织相比,癌组织中FOXC1、FOXCUT表达水平均升高(P<0.05);FOXC1、FOXCUT表达与肝细胞癌临床参数分析显示FOXC1、FOXCUT与性别、年龄、肿瘤大小差异无统计学意义(P>0.05),FOXC1、FOXCUT在有淋巴结转移、分化程度低、TNM分期为III-IV期、有肝硬化患者中表达量高于无淋巴结转移、分化程度中高、TNM分期为I-II期、无肝硬化患者(P<0.05);Pearson法分析结果显示FOXC1与FOXCUT表达水平为显著正相关;Kaplan-Meier法分析结果显示FOXC1阳性表达组3年内存活率低于阴性表达组(28.0%vs 62.0%),FOXCUT高表达组3年内存活率低于低表达组(22.0%vs 51.2%)(P<0.05)。结论:FOXC1和FOXCUT在肝细胞癌组织中高表达,二者有极大相关性,FOXC1和FOXCUT高表达不利于预后。Objective:To investigate the expressions of FOXC1 gene and upstream promoter non-coding protein FOXCUT in hepatocellular carcinoma tissues and to analyze the correlation of FOXC1 and FOXCUT.Methods:62 patients with hepatocellular carcinoma admitted to our hospital from September 2013 to June 2015 were selected as the study subjects.The expression of FOXC1 mRNA and lncRNA FOXCUT in hepatocellular carcinoma was detected by q-PCR.The expression of FOXC1 protein in clinical paraffin sections was detected by immunohistochemistry.The relationship between FOXC1,FOXCUT and clinicopathological features was analysed.The correlation was analysed by Pearson method between FOXC1 and FOXCUT.The relationship between the expression of FOXC1,FOXCUT and survival rate was analysed by Kaplan-Meier method.Results:The expression levels of FOXC1 and FOXCUT in hepatocellular carcinoma were significantly different between the two groups.Compared with the adjacent tissues,the expression levels of FOXC1 and FOXCUT in cancer tissues were increased(P<0.05).FOXC1 and FOXCUT were not significantly different from the gender,age and tumor size of hepatocellular carcinoma(P>0.05),but FOXC1 and FOXCUT had higher expression in lymph node metastasis,poor differentiation,Ⅲ-Ⅳstage and cirrhasis.The expression of FOXC1 was significantly positively correlated with the expression of FOXCUT by Pearson method,and the survival rate of FOXC1 positive expression group was lower than that of negative expression group in three years(28.0%vs 62.0%).The three-year survival rate of high FOXCUT expression group was lower than that of low FOXCUT expression group(22.0%vs 51.2%)(P<0.05).Conclusion:FOXC1 and FOXCUT are highly expressed in hepatocellular carcinoma tissues,which are highly correlated.High expression of FOXC1 and FOXCUT is not conducive to prognosis.
