摘 要:目的观察肺腺癌组织中糖原合成酶激酶(glycogen synthesis kinase,GSK)3βmRNA和蛋白的表达情况,并探讨其在肺腺癌发生发展中的意义。方法随机选取55例肺腺癌患者的癌组织标本作为实验组,相对应的癌旁正常组织标本作为对照组。用RT-PCR法检测两组中GSK3βmRNA相对表达量;用Western blot法检测两组中GSK3β蛋白的相对表达量。结果实验组GSK3βmRNA及GSK3β蛋白的相对表达量分别为(0.366±0.138)和(0.460±0.242),对照组分别为(0.485±0.137)和(0.689±0.183)。实验组GSK3βmRNA及GSK3β蛋白表达量均低于对照组(P<0.01)。肺腺癌Ⅰ~Ⅱ期患者GSK3βmRNA及蛋白相对表达量明显高于Ⅲ期患者(P<0.01)。结论相比癌旁正常组织,GSK3β在肺腺癌组织中表达下调,伴随临床分期上升,GSK3β表达下降。提示GSK3β对肺腺癌的发生发展有抑制作用,提高GSK3β的表达可能使肺腺癌的治疗获益。Objective To observe the expressions of messenger ribonucleic acid(mRNA)and protein of glycogen synthesis kinase-3 beta(GSK3β)in lung adenocarcinoma tissues,and to explore their clinical significances in the occurrence and development of lung adenocarcinoma.Methods The cancer tissue samples of 55 patients with lung adenocarcinoma were randomly selected as the experimental group and the matched normal tissue samples beside the cancer tissues as the control group.The relative expressions of GSK3βmRNA in the two groups were detected by reverse transcription-polymerase chain reaction(RT-PCR).The relative expressions of GSK3βprotein in the two groups were detected by Western blot.Results The relative expressions of GSK3βmRNA and GSK3βprotein were(0.366±0.138),(0.460±0.242)in the experimental group respectively,and(0.485±0.137)and(0.689±0.183)in the control group respectively.The expressions of GSK3βmRNA and GSK3βprotein in the experimental group were significantly lower than those in the control group(P<0.01).The relative expressions of GSK3βmRNA and GSK3βprotein in the patients with stageⅠ~Ⅱlung adenocarcinoma were significantly higher than those in the patients with stageⅢlung adenocarcinoma(P<0.01).Conclusion The expression of GSK3βis down-regulated in lung adenocarcinoma tissues compared with that in the matched normal tissues beside the cancer tissues,and the expression of GSK3βdecreased with the increase of clinical stages,suggesting that GSK3βmay inhibit the occurrence and development of lung adenocarcinoma,and that increasing the expression of GSK3βmay benefit the treatment of lung adenocarcinoma.
