摘  要:目的探讨恒清Ⅱ号方对阿尔茨海默病模型小鼠认知行为的影响。方法将72只雄性C57BL/6小鼠随机分为假手术组、模型组、恒清Ⅱ号方低剂量组、恒清Ⅱ号方中剂量组、恒清Ⅱ号方高剂量组和盐酸多奈哌齐组,每组12只。采用海马CAI区Aβ1-42寡聚体溶液注射方法造模。造模次日起,恒清Ⅱ号方低、中、高剂量组小鼠分别给予恒清Ⅱ号方20 mg/kg、40 mg/kg、80 mg/kg灌胃,盐酸多奈哌齐组给予盐酸多奈哌齐片5 mg/kg灌胃,假手术组和模型组小鼠给予生理盐水灌胃,均1次/d,连续8周。灌胃结束后采用水迷宫实验检测各组小鼠的认知行为。结果水迷宫实验第1-4天,模型组小鼠逃避潜伏期和定位航行路径长度均明显长于假手术组(P均<0.05),恒清Ⅱ号方中、高剂量组和盐酸多奈哌齐组小鼠逃避潜伏期和定位航行路径长度均明显短于模型组(P均<0.05);水迷宫实验第3天和第4天,恒清Ⅱ号方中、高剂量组和盐酸多奈哌齐组小鼠逃避潜伏期和定位航行路径长度均明显短于恒清Ⅱ号方低剂量组(P均<0.05);水迷宫实验第4天,恒清Ⅱ号方高剂量组小鼠逃避潜伏期和定位航行路径长度均明显短于恒清Ⅱ号方中剂量组和盐酸多奈哌齐组(P均<0.05)。水迷宫实验第5天,模型组小鼠穿越隐匿平台次数明显少于假手术组(P<0.05),恒清Ⅱ号方中、高剂量组和盐酸多奈哌齐组小鼠穿越隐匿平台次数均明显多于模型组和恒清Ⅱ号方低剂量组(P均<0.05),且恒清Ⅱ号方高剂量组明显多于恒清Ⅱ号方中剂量组和盐酸多奈哌齐组(P均<0.05)。结论恒清Ⅱ号方可明显改善阿尔茨海默病模型小鼠的学习记忆能力,且量效关系呈正相关,高剂量恒清Ⅱ号方的效果优于盐酸多奈哌齐。Objective It is to explore the effect of HengqingⅡdecoction on cognitive behavior in Alzheimer's disease(AD)model mice.Methods 72 male C57BL/6 mice were randomly divided into sham-operation group(sham group),model group,HengqingⅡdecoction low dose group(low group),HengqingⅡdecoction medium dose group(medium group),HengqingⅡdecoction high dose group(high group)and donepezil hydrochloride(DH)group,with 12 mice in each group.Aβ1-42 oligomer solution was injected at Hippocampal CAI area for modeling.From the next day of modeling,the mice in the HengqingⅡdecoction low,medium,and high dose groups were given 20 mg/kg,40 mg/kg,and 80 mg/kg HengqingⅡdecoction by gavage,respectively,and the DH group was given 5 mg/kg DH by gavage,sham operation group and model group were given normal saline by gavage,all once per day,for 8 consecutive weeks.After the intragastric administration,the water maze test was used to detect the cognitive behavior of mice in each group.Results From the first day to the 4th day in the water maze experiment,the escape latency and positioning path length of the mice in the model group were significantly longer than those in the sham operation group(all P<0.05),and the latency and length of the mice in the medium group,high group and DH group were significantly shorter than those in the model group(P<0.05).On the 3rd and 4th day of the water maze experiment,these indexes in the medium,high group and DH group were significantly shorter than those of the low group(all P<0.05).On the 4th day of the water maze experiment,the mice’s escape latency and positioning sailing path lengths of the high group were significantly shorter than those of the medium group and DH group(all P<0.05).On the 5th day of the water maze experiment,the number of mice crossing the hidden platform in the model group was significantly less than that in the sham operation group(P<0.05),the number of mice crossing the hidden platform in the medium group,high group and DH group were significantly more than those in the m
