摘 要:目的:探讨河南汉族人群mir-34b/c基因rs4938723多态性与结直肠癌(CRC)遗传易感性及预后的关系。方法:收集180例CRC患者术前及180例体检健康者(对照)空腹静脉血,采用PCR-RFLP法分析mir-34b/c基因启动子区rs4938723多态性;采用Kaplan-Meier法针对不同基因型患者绘制生存曲线并进行log-rank检验,采用Cox回归模型分析rs4938723多态性与CRC预后的关系。结果:CRC组C等位基因频率高于对照组(P<0.05);Kaplan-Meier生存分析结果显示,TT基因型与TC+CC基因型患者生存曲线差异有统计学意义(P<0.05),TC+CC基因型患者生存状况差于TT型患者。Cox回归分析结果显示,与TT基因型比较,TC、CC基因型是CRC预后不良的危险因素,HR(95%CI)分别为1.284(1.035~1.593)、2.042(1.415~2.946)。结论:mir-34b/c基因rs4938723多态性与河南汉族人群CRC遗传易感性及预后关系密切。Aim:To investigate the relationship between microRNA-34b/c(mir-34b/c)rs4938723 polymorphism and colorectal cancer(CRC)genetic susceptibility and prognosis in Henan Han population.Methods:A total of 180 CRC patients and 180 healthy people were enrolled in the study,and rs4938723 polymorphism in promoter region of mir-34b/c gene was analyzed by PCR-RFLP.The survival curves of patients with different genotypes were drawn by Kaplan-Meier method and were compared by log-rank test.Cox regression model was used to analyze the relationship between rs4938723 polymorphism and CRC prognosis.Results:The frequency of C allele in CRC group was higher than that in healthy group(P<0.05).Kaplan-Meier survival analysis showed that the survival curve of TT genotype patients differed from that of the TC+CC patients(P<0.05),and the survival situation of TC+CC patients was worse than that of TT patients.Cox regression analysis showed that TC,CC genotypes were the risk factors of poor prognosis of CRC,and HR(95%CI)were 1.284(1.035-1.593),2.042(1.415-2.946),respectively.Conclusion:The polymorphism of mir-34b/c rs4938723 locus is closely related to CRC genetic susceptibility and prognosis in Henan Han population.
