摘  要:从甘肃省安定区采集了4个品种的马铃薯黑痣病薯块共18个样本,采用常规组织分离法分离纯化得到98株具立枯丝核菌典型特征的菌株,经DAPI染液观察其核相,结合载玻片配对法测定其菌丝融合群。结果表明,98株立枯丝核菌均为多核,介于3~25个之间。其中,AG3融合群20株(占总菌株数20.4%),AG5融合群60株(占总菌株数61.2%),AG9融合群8株(占总菌株数8.2%),10株与标准菌株均不融合(占总菌株数10.2%)。4个品种中不同菌丝融合群分离频率有差异。AG5融合群的分离频率均最高。其中,在庄薯3号的分离频率达65.5%,为甘肃省安定区马铃薯黑痣病优势菌丝融合群,且其越冬能力强于其他菌丝融合群。A total of 18 potato tuber samples with black scurf disease were collected from four cultivars in the Anding district of Gansu province,and 98 strains with typical characteristics of Rhizoctonia solani were isolated and purified by conventional tissue isolate method.The nuclear phase was observed by the DAPI staining,and the anastomosis group was determined by the paired slide method.The results showed that the 98 isolates of Rhizoctonia solani were multinucleated,and the nuclear numbers were ranging from 3 to 25.Among them,20,60,and 8 isolates were in the AG3,AG5,and AG9 anastomosis group,accounting for 20.4%,61.2%,and 8.2%of total isolates,and 10 isolates did not fuse with standard isolates,accounting for 10.2%of the total.The isolation frequencies of different dominant anastomosis groups were different in 4 potato cultivars.The AG5 anastomosis group had the highest isolation frequency,especially in Zhuangshu No.3,which reached 65.5%.It is the dominant anastomosis group of Rhizoctonia solani in the Anding District,and its overwintering ability is stronger than other anastomosis groups.
