瑶药小钻乙醇提取物对4种肿瘤细胞增殖的影响

摘　　要：目的探讨瑶药小钻乙醇提取物对4种肿瘤细胞增殖的影响,评价其体外抗肿瘤活性。方法制备不同终浓度(200μg/mL、150μg/mL、100μg/mL、50μg/mL、25μg/mL)小钻80%乙醇提取物及顺铂。取对数生长期人乳腺癌细胞MCF-7、人结直肠癌细胞LOVO、人肝癌细胞HepG2、小鼠黑色素瘤细胞B16,采用四甲基噻唑蓝法检测不同浓度小钻乙醇提取物及顺铂作用48 h后4种肿瘤细胞的增殖情况,计算增殖抑制率及半数抑制浓度(IC 50)。结果各浓度小钻乙醇提取物对4种肿瘤细胞增殖均有一定抑制作用,抑制率随浓度的增加而升高(均P<0.05),但终浓度最高为200μg/mL时抑制率<80%。小钻乙醇提取物对4种细胞的IC 50值均>20μg/mL,而顺铂的IC 50值均<10μg/mL,小钻乙醇提取物对4种细胞的抑制作用均低于顺铂(均P<0.05)。结论小钻乙醇提取物对4种肿瘤细胞的体外增殖有一定的抑制作用,但其致死作用不明显,活性低于顺铂。Objective To explore the effect of ethanol extract from Yao medicine Kadsura longipedunculata Finet et Gagnep.on the proliferation of four types of tumor cells,thus evaluating its in vitro anti-tumor activities.Methods Cisplatin and 80%ethanol extract from Kadsura longipedunculata Finet et Gagnep.of various final concentrations(200μg/mL,150μg/mL,100μg/mL,50μg/mL and 25μg/mL)were prepared.Human breast cancer cells MCF-7,human colorectal tumor cells LOVO,human liver hepatocellular carcinoma cells HepG2,and mouse melanoma cells B16 at logarithmic growth phase were collected,methyl thiazolyl tetrazolium assay was employed to determine the proliferation of four types of tumor cell strains after 48 hours of intervention by cisplatin or ethanol extract from Kadsura longipedunculata Finet et Gagnep.of various final concentrations,then the proliferation inhibition rate and half maximal inhibitory concentration(IC 50)were calculated.Results Ethanol extract from Kadsura longipedunculata Finet et Gagnep.of various final concentrations exerted a certain inhibitory effect on four types of tumor cell strains,and the inhibitory rate rose with the increase of concentration(all P<0.05);however,the inhibitory rate was less than 80%at the highest final concentration of 200μg/mL.IC 50 values of ethanol extract from Kadsura longipedunculata Finet et Gagnep.on the four types of tumor cell strains were all higher than 20μg/mL,whereas IC 50 values of cisplatin were lower than 10μg/mL,the inhibitory effect of ethanol extract from Kadsura longipedunculata Finet et Gagnep.on the four types of tumor cell strains was milder than that of cisplatin(all P<0.05).Conclusion Ethanol extract from Kadsura longipedunculata Finet et Gagnep.,in some degree,possesses an inhibitory effect on the proliferation of four types of tumor cells in vitro,but its lethal effect is unobvious,and its activity is lower than that of cisplatin.
