摘  要:目的:研究表明骨髓间充质干细胞(MSC)的功能改变在骨髓增生异常综合征(MDS)的造血功能衰竭过程中发挥了重要的作用,但其作用机制有待进一步阐明。因此,本研究对MDS患者的MSC中参与骨髓形成的基因Runx2和Osterix表达特点进行分析。方法:以在我院就诊的MDS患者为研究对象,通过细胞形态、免疫表型和成骨细胞、脂肪细胞分化潜能对体外培养的MSC进行鉴定,q-PCR对MSC中Runx2和Osterix基因表达进行分析,以营养性贫血患者的MSC作为对照组。结果:MDS患者的MSC和对照组MSC均表达相似的免疫表型和具有向成骨细胞、脂肪细胞分化的潜能。然而,MDS患者的MSC中Osterix基因的表达水平显著低于对照组(P<0.01);MDS患者MSC中Runx2的表达水平亦显著低于对照组(P<0.05)。结论:MDS患者的MSC中参与骨髓形成的Osterix和Runx2基因表达水平降低。Objective:Studies have shown that changes in the function of bone marrow mesenchymal stem cells(MSCs) play an important role in the process of hematopoietic failure in myelodysplastic syndromes(MDS).However,the mechanism of action needs to be further elucidated.Therefore,this study analyzed the expression characteristics of Runx2 and Osterix genes involved in bone marrow formation in MSCs of MDS patients.Method:MDS patients treated in our hospital were used as the research objects.The MSCs were identified by cell morphology,immunophenotype and differentiation potential of osteoblasts and adipocytes in vitro.The expression of Runx2 and Osterix in MSCs was analyzed by q-PCR,and the MSCs of nutritional anemia patients were used as the control group.Result:The MSCs of patients with MDS and the MSCs of control group both expressed similar immunophenotype and had the potential to differentiate into osteoblasts and adipocytes.However,the expression level of Osterix gene in MSCs of MDS patients was significantly lower than that in the control group(P<0.01);the expression level of Runx2 in MSCs of MDS patients was significantly lower than that in the control group(P<0.05).Conclusion:The expression of Osterix and Runx2 genes involved in bone marrow formation in MSCs of MDS patients decreased.
