摘 要:国标法和新型试剂盒法利用植物乳杆菌对生物素的高度特异性和敏感性,定量测出乳粉样品中生物素的含量。通过对标准乳粉质控样品的检测,测得国标法标准曲线在0.00~1.00 ng范围内线性关系良好(R^2=0.9990),方法检出限为2.0μg/100 g,所测样品RSD为3.22%(n=6);测得试剂盒法标准曲线在0.00~0.52μg/100 g范围内线性关系良好(R^2=0.9923),方法检出限为0.02μg/100 g,所测样品RSD为3.18%(n=6);两种方法生物素测定值均在给定值范围内,且差异不显著(p>0.05)。在50%,100%和200%添加水平下,生物素回收率均在90%~110%之间(RSD<10%),样品重复性试验结果RSD小于5%。两种方法对标准质控样检测结果稳定准确、重复性好,适用于乳粉中生物素含量的检测。According to the characteristics of sensitivity and specificity to the free biotin by Lactobacillus plantarum,the biotin content in standard quality milk powder was determined by national standard method and the novel kit method.The result showed that the national standard method linear relationship for the standard curve in the range of 0.00-1.00 ng was good(R^2=0.9990),the detection limit was 2.0μg/100 g,and the standard deviation(RSD)was 3.22%(n=6).The kit method linear relationship for the standard curve in the range of 0.00-0.52μg/100 g was good(R^2=0.9923)too,the detection limit was 0.02μg/100 g,and the standard deviation(RSD)was 3.18%(n=6).The content of biotin tested by the two methods had no significant difference(p>0.05),and they all conformed to the scope of the limited range.The recovery rates were all 90%-110%(RSD<10%)under adding level 50%,100%and 200%.The RSD of the sample repeatability test results was less than 5%.The two methods can be widely applied to the detection of biotin in milk powder.
