摘 要:为了解安徽省参加品比试验的各茶区茶树品系的遗传多样性,选取50对SSR引物对安徽省各茶区68份茶树品系进行分析,共检测出650个等位基因,平均每对引物的等位基因数为13个,50对引物扩增的片段长度在95~370 bp,有效等位基因(Ne)值为1.05~17.22,平均为6.10;观测杂合度(Ho)值为0.00~0.99,平均为0.38;期望杂合度(He)值为0~1,平均为0.62;各引物的Shannon信息指数(I)值为0.12~3.06,平均为1.86;多态信息含量(PIC)值变化范围在0.04~0.94,平均为0.72;基因流(Nm)值为0.11~4.45,平均为1.03。选出8对核心引物组合,构建DNA指纹图谱。聚类结果表明,68份茶树品系的居群可聚成3类,与居群的地理分布有一定的相似性,地理距离越近的品系,遗传一致度越大。利用SSR分子标记技术可以为安徽省茶树良种的改良和保护提供参考。In order to understand the genetic diversity of tea strain in different tea areas of Anhui Province, A total of 650 alleles were detected with selected 50 pairs of SSR primers for analysis of 68 tea tree strains. The average number of alleles of each pair of primers was 13. The length of fragment amplified by 50 pairs of primers was95~370 bp. The effective allele(Ne) value was 1.05~17.22, with an average of 6.10. The observed heterozygosity(Ho) value was 0.00~0.99, with an average of 0.38. The expected heterozygosity(He) value was 0~1, with an average of 0.62. The Shannon information index(I) value of each primer was 0.12~3.06, with an average of 1.86.The polymorphic information content(PIC) ranged from 0.04~0.94, with an average of 0.72. Gene flow(Nm)values ranged from 0.11~4.45, with an average of 1.03. Eight pairs of core primer combinations were selected to construct DNA fingerprint. The clustering results showed that the populations of 68 tea tree strains can be clustered into 3 groups, which had a certain similarity with the geographical distribution of the populations. The closer geographical distance, the greater genetic consistency. SSR molecular marker technology can provide reference for the improvement and protection of tea tree varieties in Anhui Province.
