摘  要:目的探讨杨梅素对环磷酰胺致小鼠肝损伤的保护作用及其可能的作用机制。方法采用连续7d腹腔注射环磷酰胺(50 mg/kg)建立小鼠肝损伤模型,随后连续30 d分别灌胃杨梅素(100、200、400 mg/kg)进行药物干预。取材后计算小鼠的肝指数;检测各组小鼠血清中血清谷丙转氨酶(ALT)和谷草转氨酶(AST)活性;肝组织中还原型谷胱甘肽(GSH)、丙二醛(MDA)及一氧化氮(NO)含量;酶联免疫吸附法(ELISA)检测肝组织匀浆中肿瘤坏死因子-α(TNF-α)、核因子kb(NF-kb)、白介素-1β(IL-1β)的表达情况;蛋白质免疫印迹法(Western blot,WB)检测肝组织Bax、Caspase-3蛋白的表达。结果与模型组比较,杨梅素中、高剂量组肝指数降低,血清AST、ALT活性降低,肝组织中GSH含量增加,MDA、TNF-α、NF-kb、IL-1β、NO含量均不同程度降低,WB结果显示Bax、caspase-3表达显著下调。结论杨梅素对环磷酰胺诱导小鼠肝损伤有一定保护作用,其机制可能与抗氧化应激、抗炎和抗肝细胞凋亡有关。Objective To investigate the protective effect of myricetin on liver injury induced by cyclophosphamide in mice and its possible mechanism.Methods Liver injury model was established in mice via intraperitoneal injection of cyclophosphamide(50 mg/kg)for 7 days.Mice in the treatment groups were treated with myricetin at 100 mg/kg/ip,200 mg/kg/ip and 400 mg/kg/ip for 30 days,respectively.The mice were sacrificed on day 31.Liver index was calculated after sampling.Levels of alanine aminotransferase(ALT)and aminotransferase(AST)in serum,as well as glutathione(GSH),malondialdehyde(MDA)and nitric oxide(NO)in liver hemogenates were assayed.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of tumor necrosis factor-alpha(TNF-α),nuclear factor KB(NF-kb)and interleukin-1 beta(IL-1β)in liver homogenates.The expressions of Bax and caspase-3 in liver tissues were detected by Western blot(WB).Results Compared with the model group,the liver index,serum AST and ALT activities were decreased.GSH content was increased,and MDA,TNF-α,NF-kb,IL-1βand NO contents in liver tissues were all decreased in different doses myricetin groups.WB results showed that Bax and caspase-3 expressions were significantly down-regulated in the 200 mg/kg,400 mg/kg myricetin groups.Conclusion Myricetin has a protective effect on liver injury induced by cyclophosphamide in mice,and its mechanism may be related to anti-oxidative stress,anti-inflammation and anti-apoptosis.
