摘  要:目的研究有丝分裂原活化蛋白激酶激酶激酶3(mitogen-activated protein kinase kinase kinase 3,MEKK3)与宫颈癌患者凋亡抑制基因B淋巴细胞瘤-2基因(B lymphocytoma-2 gene,bcl-2)表达的相关性。方法选取2017年1月至2018年12月收治的宫颈癌患者30例,取所有宫颈癌患者手术切除癌组织及癌旁组织标本,行免疫组化染色,检测宫颈癌癌组织及癌旁组织中MEKK3、bcl-2的表达,并分析MEKK3、bcl-2与宫颈癌患者临床病理特征的关系,采用Pearson分析MEKK3、bcl-2之间的相关性。结果宫颈癌癌组织中MEKK3、bcl-2阳性表达率均高于癌旁组织,差异有统计学意义(P<0.05)。MEKK3、bcl-2与宫颈癌患者年龄、组织学类型无关,差异无统计学意义(P>0.05);MEKK3、bcl-2与宫颈癌患者FIGO分期、肌层浸润、淋巴结转移、分化程度有关,MEKK3、bcl-2在Ⅲ+Ⅳ期、肌层浸润>1/2、有淋巴结转移、中-高分化的宫颈癌患者中阳性表达率高于在Ⅰ期、Ⅱ期、肌层浸润≤1/2、无淋巴结转移、低分化的宫颈癌患者中阳性表达率,差异有统计学意义(P<0.05)。MEKK3、bcl-2之间呈现正相关(r=0.521,P=0.003)。结论MEKK3在宫颈癌中呈现高表达,且与凋亡抑制基因bcl-2之间具有一定的相关性,两者在宫颈癌的发生发展中具有协同作用。Objective To investigate the correlation between mitogen-activated protein kinase kinase kinase 3(MEKK3)and the expression of B lymphocytoma-2 gene(bcl-2)in patients with cervical cancer.Methods Thirty patients with cervical cancer who were admitted and treated in our hospital from January 2017 to December 2018 were enrolled in the study.The surgical ablation specimens of cervical cancer tissues and adjacent tissues were taken,and the expression levels of MEKK3 and Bcl-2 in cervical cancer tissues and adjacent tissues wee detected by immunohistochemical staining.Moreover the correlation between the expression levels of MEKK3,Bcl-2 and clinicopathological characteristics of cervical cancer was analyzed.In additon the correlation between MEK3 and bcl-2 was analyzed by means of Pearson analysis.Results The positive expression rates of MEKK3 and Bcl-2 in cervical cancer tissues were significantly higher than those in adjacent tissues(P<0.05).MEKKK3 and Bcl-2 were not related to patient’s age and histological type of cervical cancer(P>0.05),however,which were closely related to FIGO stage,myometrial invasion,lymph node metastasis and differentiation of cervical cancer.The positive expression rates of MEKK3 and Bcl-2 in patients at stageⅢ+Ⅳ,with myometrial invasion>1/2,lymph node metastasis and moderate-high differentiation cervical cancer were significantly higher than those of patients at stageⅠ,Ⅱand myometrial differentiation.There were significant differences in the positive expression rates in cervical cancer patients with layer infiltration(<1/2),no lymph node metastasis and poor differentiation(P<0.05).Moreover there was a positive correlation between MEKK3 and Bcl-2(P<0.01).Conclusion MEKK3 is highly expressed in cervical cancer,which has a certain correlation with apoptosis suppressor gene bcl-2,the two factors have a synergistic effect in the pathogenesis and development of cervical cancer.
