摘 要:目的研究肺腺癌转移相关转录本1(MALAT1)对喉癌(LC)细胞增殖和侵袭的作用及其机制。方法用实时荧光定量聚合酶链反应(qRT-PCR)检测LC细胞中MALAT1、微小RNA-503-5p(miR-503-5p)和叉头盒蛋白K1(FOXK1)的表达水平;用生物学软件预测MALAT1、miR-503-5p和FOXK1的靶向关系,双荧光素酶报告进一步验证;蛋白质印迹技术(Western blot)检测相关蛋白的表达水平,细胞计数试剂盒8(CCK8)、侵袭小室(Transwell)和克隆形成检测LC细胞的增殖、侵袭情况。结果在LC细胞中,MALAT1和FOXK1高表达,miR-503-5p低表达;且miR-503-5p是MALAT1的靶向调节基因之一,两者呈负相关;下调miR-503-5p可以明显促进LC细胞的增殖和侵袭(P<0.01),并逆转MALAT1低表达对LC细胞增殖和侵袭的抑制作用。FOXK1是miR-503-5p的靶向调节基因之一,过表达FOXK1明显促进LC细胞的增殖和侵袭能力,且逆转MALAT1低表达对LC细胞增殖和侵袭的抑制作用。结论MALAT1通过靶向miR-503-5p上调FOXK1促进LC细胞的增殖和侵袭能力。Objective To study the effect of metastasis-associated lung adenocarcinoma transcript 1(MALAT1)on the proliferation and invasion of laryngeal cancer(LC)cells and its mechanism.Methods qRT-PCR was used to detect the expression levels of MALAT1,micro RNA-503-5p(miR-503-5p)and forkhead box M1(FOXK1)in LC cells.The biological software was used to predict the target relationship of MALAT1,miR-503-5p and FOXK1 which was further verified by Dual Luciferase Reporter.Western blot was applied to detect the protein expression.CCK8,Transwell and colony formation was employed to detect the proliferation and invasion of LC cells.Results In LC cells,MALAT1 and FOXK1 were highly expressed,while miR-503-5p was lowly expressed.miR-503-5p was one of the targeted regulatory genes of MALAT1 and negatively regulated by MALAT1.The down-regulation of miR-503-5p significantly promoted the proliferation and invasion ability of LC cells(P<0.01)and reversed the inhibitory effect of the low expression of MALAT1 on the proliferation and invasion of LC cells.It was also demonstrated that FOXK1 was one of the targeted regulatory genes of miR-503-5p.The overexpression of FOXK1 significantly promoted the proliferation and invasion of LC cells and reversed the inhibitory effect of the low expression of MALAT1 on the proliferation and invasion of LC cells.Conclusion MALAT1 promotes the proliferation and invasion of LC cells by targeting miR-503-5p to up-regulate FOXK1.
