摘  要:为了考察抗阻运动对海洛因诱导大鼠的自身给药及伏隔核基因表达的影响,本研究将40只雌性SD大鼠随机分为抗阻运动组(n=20)和安静对照组(n=20)。抗阻运动组大鼠进行4周的负重爬梯训练。采用q RT-PCR和Western blotting检查两组大鼠药物成瘾相关基因的mRNA和蛋白表达。结果表明,抗阻运动组大鼠(海洛因剂量为0.001 mg/kg,0.005 mg/kg和0.01 mg/kg)的自身给药输注次数明显低于安静组;抗阻运动组大鼠NAc核区中的阿片样物质受体(OPRK1(Opioid receptor kappa1),Oprm1(Opioid receptor mu1))和多巴胺受体(D1,D2,D3)的mRNA和蛋白表达明显低于安静组大鼠,而脑源性神经营养因子外显子(Ⅰ,ⅡA,ⅡB)的mRNA和蛋白表达均显著高于安静组;两组大鼠NAc壳区的上述基因表达无显著差异。本研究发现抗阻运动减少了海洛因诱导大鼠的自身给药,并引起了大鼠药物成瘾性相关基因表达的变化。In order to investigate the effects of resistance exercise on self-administration and nucleus accumbens gene expression in heroin-induced rats.In this study,40 female SD rats were randomly divided into the resistance exercise group(n=20)and the quiet control group(n=20).Rats in the resistance exercise group were subjected to4-week load-lifting training.The mRNA and protein expressions of drug addiction-related genes in the two groups were examined by q RT-PCR and Western blotting.The results showed that the number of self-administered infusions in the resistance exercise group(heroin doses were 0.001 mg/kg,0.005 mg/kg,and 0.01 mg/kg)was significantly lower than that of quiet group.The mRNA and protein expression of opioid receptors(OPRK1(Opioid receptor kappa1)and Oprm1(Opioid receptor mu 1))and dopamine receptors(D1,D2,D3)in the NAc core of the resistance exercise group were significantly lower than that of the quiet group.The m RNA and protein expression of brain-derived neurotrophic factor exons(Ⅰ,ⅡA,ⅡB)were significantly higher in the resistance exercise group than in the quiet group.There was no significant difference in the expression of the above genes in the NAc shell between the two groups.In this study,we found that resistance exercise reduces self-administration of heroininduced rats and causes changes in the expression of drug addiction-related genes in rats.
