摘  要:为了分析阿替普酶联合丁苯酞对缺血性脑卒中大鼠的保护机制,为阿替普酶和丁苯酞的临床应用提供基础研究,本研究随机将Sprague-Dawley(SD)大鼠分为假手术组、模型组、阿替普酶组、丁苯酞组和阿替普酶+丁苯酞组,每组30只;采用线栓法制作脑缺血脑卒中大鼠模型,后3组分别给药后统计分析各组大鼠脑组织含水量、炎症因子、血管内皮生长因子(vascular endothelial growth factor,VEGF)蛋白表达、神经和认知功能水平。与假手术组大鼠相比,模型组缺血性脑卒中大鼠脑组织含水量和炎症因子水平明显升高(p<0.05),而VEGF蛋白表达、神经和认知功能水平明显降低(p<0.05)。与模型组大鼠相比,阿替普酶和丁苯酞均明显降低缺血性脑卒中大鼠脑组织含水量和炎症因子水平(p<0.05),升高VEGF蛋白表达水平、改善神经和认知功能障碍(p<0.05),但是两者联合使用的疗效比单独使用阿替普酶和丁苯酞更强。本研究结果表明,阿替普酶和丁苯酞均能有效改善脑缺血性脑卒中,但是两者联合使用的疗效更好,值得临床使用和推广。To analyze the protective mechanism of alteplase combined with butylphthalide in rats with ischemic stroke,and to provide basic research for the clinical application of alteplase and butylphthalide,in this study,Sprague-Dawley(SD)rats were randomly divided into sham operation group,model group,ateplase group,butylphthalide group,ateplase+butylphthalide group,with 30 rats in each group.Rat models of cerebral ischemia and stroke were made by bolt-line method.After administration of the drugs,brain tissue water content,inflammatory factors,expression of Vascular endothelial growth factor(VEGF),neurological and cognitive function were analyzed.Compared with sham-operated rats,the water content and inflammatory factors in brain tissue of ischemic stroke rats in model group increased significantly(p<0.05),while the expression of vascular endothelial growth factor protein,neurological and cognitive function decreased significantly(p<0.05).Compared with the model group,both ateplase and butylphthalide significantly decreased the water content and inflammatory factors in cerebral tissue,increased the expression of vascular endothelial growth factor and improved neurological and cognitive impairment in rats with ischemic stroke(p<0.05),increased the expression level of VEGF protein,and improved neurological and cognitive dysfunction(p<0.05).The results of this study show that both alteplase and butylphthalide can effectively improve ischemic stroke,but the combination of the two is more effective,worthy of clinical use and promotion.
