摘 要:目的探讨槲皮素对人宫颈癌HeLa细胞凋亡及迁移的影响,并探讨其可能的机制。方法以不同浓度槲皮素(0,20,40,80μmol/L)处理生长状态良好的HeLa细胞48 h,用流式细胞术检测细胞凋亡,Transwell检测细胞迁移,流式细胞术检测细胞中活性氧(ROS)含量,ELISA法检测细胞丙二醛(MDA)含量及总超氧化物歧化酶(T-SOD)活力。结果0,20,40,80μmol/L槲皮素组细胞凋亡率依次分别为(4.61±0.76)%,(9.47±0.54)%,(14.78±0.93)%,(23.34±1.96)%;四组之间两两比较,差异均有统计学意义(均P<0.05)。0,20,40,80μmol/L槲皮素组细胞迁移数依次分别为(104.67±5.03)个,(94.00±3.46)个,(75.00±9.54)个,(55.67±4.62)个;四组之间两两比较,差异均有统计学意义(均P<0.05)。0,20,40,80μmol/L槲皮素组ROS表达量依次分别为65402.00±886.87,76497.00±9648.64,95774.66±3440.82,106135.00±3724.35;四组之间两两比较,差异均有统计学意义(均P<0.05)。0,20,40,80μmol/L槲皮素组MDA含量依次分别为(1.85±0.06)nmol/mg,(2.52±0.22)nmol/mg,(3.50±0.24)nmol/mg,(4.19±0.43)nmol/mg;四组之间两两比较,差异均有统计学意义(均P<0.05)。0,20,40,80μmol/L槲皮素组T-SOD活力依次分别为(75.28±0.39)U/mg,(62.84±3.76)U/mg,(52.61±1.51)U/mg,(35.52±3.72)U/mg;四组之间两两比较,差异均有统计学意义(均P<0.05)。结论不同浓度槲皮素均可诱导HeLa细胞凋亡,抑制细胞迁移,增加细胞ROS及MDA含量,抑制T-SOD活力,且以上效应随着槲皮素浓度的增加而增强。槲皮素能发挥促HeLa细胞凋亡、抑制迁移的作用,可能与其增强细胞的氧化应激有关。Objective To investigate the effect of quercetin on the apoptosis and the migration of human cervical cancer HeLa cells,and to explore its possible mechanism.Methods HeLa cells in good culture condition were treated with different concentrations of quercetin(0,20,40,80μmol/L)for 48 h.Cell apoptosis was detected by flow cytometry and the migration was detected by Transwell.Reactive oxygen species(ROS)content in HeLa cells was detected by flow cytometry,and malondialdehyde(MDA)content as well as total superoxide dismutase(T-SOD)activity were detected by biological kits.Results The cell apoptosis rate was(4.61±0.76)%,(9.47±0.54)%,(14.78±0.93)%,(23.34±1.96)%in 0,20,40,80μmol/L quercetin groups,respectively,and the difference between two groups was statistically significant(all P<0.05).The number of migrated cells was 104.67±5.03,94.00±3.46,75.00±9.54,55.67±4.62 in 0,20,40,80μmol/L quercetin groups,respectively,and the difference between two groups was statistically significant(all P<0.05).The expression of ROS was 65402.00±886.87,76497.00±9648.64,95774.66±3440.82,106135.00±3724.35 in 0,20,40,80μmol/L quercetin groups,respectively,and the difference between two groups was statistically significant(all P<0.05).The expression of MDA was(1.85±0.06)nmol/mg,(2.52±0.22)nmol/mg,(3.50±0.24)nmol/mg,(4.19±0.43)nmol/mg in 0,20,40,80μmol/L quercetin groups,respectively,and the difference between two groups was statistically significant(all P<0.05).The activity of T-SOD was(75.28±0.39)U/mg,(62.84±3.76)U/mg,(52.61±1.51)U/mg,(35.52±3.72)U/mg in 0,20,40,80μmol/L quercetin groups,respectively,and the difference between two groups was statistically significant(all P<0.05).Conclusion Different concentrations of quercetin could induce HeLa cell apoptosis,inhibit cell migration,increase ROS and MDA contents,and inhibit T-SOD activity,and the above effects of quercetin are enhanced with the increase of its concentration.Quercetin can promote the apoptosis and inhibit the migration of HeLa cells,which may be relate
