摘  要:目的观察下调CD133基因表达对乳腺癌细胞MDA-MB-231侵袭和迁移的影响,并从上皮间质转化方向研究侵袭和迁移的机制。方法采用Western blot方法检测正常人乳腺细胞MCF-10A和乳腺癌细胞株MDA-MB-231、SKBr3、MCF-7中CD133的表达,以及正常乳腺组织和乳腺癌组织中CD133的表达。以CD133高表达的乳腺癌细胞株MDA-MB-231为后续实验对象,实验分为control组(只转染Lipofectamine 2000)、NC组(Lipofectamine 2000和NC序列)和si-CD133组(Lipofectamine 2000和si-CD133序列)。采用Transwell小室检测CD133基因下调对MDA-MB-231细胞侵袭和迁移的影响;荧光定量PCR和Western blot检测了E-cadherin、vimentin、N-cadherin的相对表达水平。结果CD133在乳腺癌细胞MDA-MB-231中的表达水平明显高于正常人乳腺细胞(P<0.05),CD133在乳腺癌组织中的表达水平明显高于正常人乳腺组织(P<0.05)。下调CD133,MDA-MB-231细胞的侵袭和迁移显著被抑制(P<0.05),波形蛋白、N-钙黏蛋白表达降低,E-钙黏蛋白表达上调(均P<0.05)。结论CD133的表达可促进乳腺癌细胞MDA-MB-231的侵袭和迁移,其机制可能与上皮间质的转化有关。Objective To investigate the effect of down-regulating the CD133 gene expression on the invasion and migration of breast cancer cells MDA-MB-231,and explore its mechanism from the epithelial-mesenchymal transition.Methods The expression of CD133 was detected in normal human breast cells MCF-10A and breast cancer cell lines MDA-MB-231,SKBr3,MCF-7,and in normal breast tissues and breast cancer tissues by Western blot.Breast cancer cell line MDA-MB-231 with high expression of CD133 was used as the experimental object,and divided into control group(transfection with Lipofectamine 2000),NC group(transfection with Lipofectamine 2000 and NC sequence)and si-CD133 group(transfection with Lipofectamine 2000 and si-CD133 sequence).Transwell chamber was used to detect the effect of down-regulation of CD133 gene on the invasion and migration of MDA-MB-231 cells.The relative expression levels of N-cadherin,E-cadherin and vimentin were detected by fluorescence quantitative PCR and Western blot.Results The expression level of CD133 in breast cancer cells was significantly higher than that in normal breast cells(P<0.05).The expression level of CD133 in breast cancer tissues was significantly lower than that in normal human breast tissues(P<0.05).When CD133 expression was down-regulated,the invasion and migration of MDA-MB-231 cells were significantly inhibited(P<0.05),vimentin and N-cadherin expression was decreased,while E-cadherin expression was up-regulated(all P<0.05).Conclusion The expression of CD133 can promote the invasion and migration of MDA-MB-231 cells,which may be related to the transformation of epithelium.
