摘  要:目的探讨A血型抗原减弱标本的血清学和分子机制研究。方法采用血清学凝集试管法检测ABO血型,再对标本进行聚合酶链反应和ABO基因6、7外显子及部分内含子的基因测序分析。结果5例标本ABO血型检测正反定型不一致,正定型表现为A抗原减弱,与抗-A1均不凝集,反定型与Bc均4+凝集,与Oc均不凝集。血清学结果A3亚型2例、A2亚型2例、Ael亚型1例,基因测序结果分别为A307/O022例、O02/O02、A102/O01、A102/A101各1例。结论A亚型的基因遗传背景复杂,血清学表现为亚型,基因存在异质性。Objective To study the serological and molecular mechanism of A antigen-attenuated specimens.Methods The ABO blood group was detected by serological agglutination test tube.Then specimens were subjected to polymerase chain reaction and sequence analysis of ABO gene 6,7 exons and some introns.Results The positive stereotypes of 5 specimens showed that the A antigen were all weakened,and were not agglutinated with anti-A1 reagent,but were both 4+agglutinated with Bc,and were not agglutinated with Oc.The results of serological tests were 2 cases of A3 subtype,2 cases of A2 subtype,and 1 case of Ael subtype.The results of gene sequencing were 2 cases of A307/O02,1 case of O02/O02,A102/O01 and A102/A101,Conclusion The genetic background of the subtype A is complex,the serology is subtype,and the gene is heterogeneous.
