摘　　要：目的:研究人白细胞相关免疫球蛋白样受体-1(LAIR-1)对卵巢癌细胞HO8910线粒体能量代谢的影响。方法:利用低氧模拟剂CoCl2处理细胞,分析缺氧对LAIR-1表达的影响;利用LAIR-1慢病毒RNAi干扰载体(LAIR-1-RNAi-lentivirus),下调LAIR-1在卵巢癌HO8910细胞中的表达。Western blot、qRT-PCR法鉴定HO8910慢病毒感染后LAIR-1的表达水平;通过CCK-8实验和细胞克隆形成实验,验证下调LAIR-1表达对HO8910细胞生长的影响;通过Seahorse生物能量分析仪,检测下调LAIR-1表达后对HO8910细胞生物能量代谢的影响。结果:在低氧环境下,HO8910细胞的LAIR-1表达显著上调;LAIR-1-RNAi-lentivirus转染后,HO8910细胞的LAIR-1表达显著下调,细胞生长能力明显增强(P<0.01);LAIR-1表达显著下调的HO8910细胞,ATP产生明显改善(P<0.001),同时其基础呼吸(P<0.001)和最大呼吸能力(P<0.001)均明显提高。结论:LAIR-1分子可能通过影响线粒体生物能量代谢抑制人卵巢癌HO8910细胞的生长。Objective:To investigate the effects of leukocyte-associated immunoglobulin-like receptor-1(LAIR-1)on mitochondrial bioenergetics of ovarian cancer cell HO8910.Methods:The expression of LAIR-1 in the tumor hypoxic condition was investigated using a chemical hypoxic model with CoCl2.The expression of LAIR-1 in ovarian cancer HO8910 cells was knocked down by transfection with LAIR-1-RNAi lentivirus.The efficiency of LAIR-1 knockdown was identified by Western blot and qRT-PCR.The cell growth ability was detected by using CCK-8 kit and colony formation assay.The Seahorse XF96 Extracellular Flux Analyzer was used to detect the effects of LAIR-1 on mitochondrial bioenergetics.Results:The expression of LAIR-1 was significantly increased in a time and dose-dependent manner in HO8910 cells under the hypoxic condition.The down-regulation of LAIR-1 in HO8910 cells promoted the ability of cell proliferation(P<0.01)and colony formation(P<0.01),and improved the production of ATP(P<0.001),basal respiration(P<0.001)and maximum respiration capacity(P<0.001).Conclusion:LAIR-1 may inhibit the proliferation of ovarian cancer HO8910 cells by affecting the mitochondrial bioenergetics.
