摘 要:目的:通过检测上皮性卵巢癌(EOC)组织中ERH蛋白表达水平,探讨其作为EOC的治疗靶点和预后生物标记物的可能性。方法:选取2019年1月至2019年3月青岛大学附属医院收治的47例EOC组织、10例正常卵巢组织和10例正常输卵管组织的手术标本,免疫组化法检测EOC、正常卵巢组织和正常输卵管组织中ERH蛋白表达水平,Western blot法和qRT-PCR法验证组织中ERH蛋白和mRNA表达水平。分析ERH表达水平与患者的年龄、临床分期(FIGO分期)、病理分级、组织学类型、淋巴结转移和卵巢癌分型的关系。Transwell实验验证干扰ERH表达对卵巢癌细胞的迁移和侵袭能力的影响。结果:EOC组织中ERH蛋白和mRNA表达明显高于正常卵巢组织和正常输卵管组织(P<0.05)。EOC组织中ERH蛋白高表达与病理分级、淋巴结转移、卵巢癌分型有关(P<0.05)。干扰卵巢癌细胞A2780中ERH表达显著降低了细胞的迁移和侵袭能力(P<0.05)。结论:ERH蛋白在卵巢癌组织中高表达,干扰卵巢癌细胞的ERH表达可抑制卵巢癌细胞的侵袭和迁移。Objective:To detect the expression level of ERH protein in epithelial ovarian cancer(EOC)tissues.To explore its potential as a therapeutic target and prognostic biomarker for EOC.Methods:47 cases of EOC tissues,10 normalovary tissues and 10 normal fallopian tubes were collected from the Affiliated Hospital of Qingdao University from January to March 2019.These tissue specimens were detected by immunohistochemical staining for expression of ERH protein.The expression of ERH at the protein and mRNA levels was verified by Western blot and qRT-PCR.The relationship between ERH expression level and patient age,clinical stage(FIGO stage),pathological grade,histological type,lymph node metastasis,and ovarian cancer classification was analyzed.Transwell experiments were used to verify that interference with ERH expression can inhibit the migration and invasion of ovarian cancer cells.Result:The protein and mRNA expression of ERH in EOC tissues were significantly higher than those in normalovary and normal fallopian tubes tissues(P<0.05).The high expression of ERH protein levels was significantly associated with pathological grade,lymph node metastasis and ovarian cancer subtype(P<0.05).Interfering with ERH expression of ovarian cancer cell A2780 significantly reduced cell migration and invasion ability(P<0.05).Conclusions:ERH protein is highly expressed in ovarian cancer,and interference with ERH expression can inhibit invasion and metastasis of ovarian cancer cells.
