摘 要:目的本研究的目的是利用化学和生物信息学的方法寻找肾癌和癌旁组织的差异蛋白,探讨目标蛋白作为肾癌生物标志物的可能性。方法采用液相色谱-质谱法鉴定肾癌和癌旁组织差异表达的蛋白,借助免疫组织化学的方法观察肾癌和癌旁组织标本中synemin蛋白的表达。结果肾癌与癌旁组织差异表达的蛋白有26个,肾癌组织中过表达18个,低表达8个。synemin蛋白在肾癌组织中的高表达,统计学差异明显(P<0.05)。结论液相色谱-质谱技术可以用于检测肾癌与癌旁组织中差异表达的蛋白,synemin蛋白具有成为肾癌生物标志物的潜力。Objective The purpose of this study was to use chemical and bioinformatics methods to find differential protein in renal cancer and adjacent tissues,and to explore the possibility of target proteins as renal cancer biomarkers.Methods The differentially expressed protein of renal cancer and adjacent tissues were identified by liquid chromatography-mass spectrometry.The expression of synemin in renal cancer and adjacent tissues was observed by immunohistochemistry.Results There were 26 proteins differentially expressed in renal cancer and adjacent tissues,18 were overexpressed and 8 were under-expressed.The expression level of synemin in renal cancer tissues was higher than that in adjacent tissues,which was statistically different(P<0.05).Conclusion We believe that liquid chromatography-mass spectrometry can be used to detect differentially expressed protein in renal cancer and adjacent tissues.Synemin has the potential to become a biomarker for renal cancer.
