摘　　要：目的:探讨磷酸化磷酸组氨酸无机焦磷酸酶(LHPP)对上皮性卵巢癌恶性行为的影响及其机制。方法:选择2016年9月至2019年9月在青岛市市立医院留存的上皮性卵巢癌及正常卵巢的新鲜组织和石蜡切片,采用免疫组化法检测LHPP在上皮性卵巢癌和正常卵巢组织中的表达;应用CCK-8增殖实验和平板克隆实验检测LHPP对卵巢癌细胞增殖能力的影响;Transwell小室实验检测对细胞侵袭、迁移能力的影响;蛋白印迹法检测上皮间质转化(EMT)相关生物标志物的表达。结果:LHPP在卵巢癌组织中的表达水平显著高于正常卵巢组织(P<0.01),上皮性卵巢癌中LHPP蛋白的表达水平与手术病理分期、组织学分级、肿瘤糖类抗原125(CA125)水平明显相关(P<0.05),而与患者年龄是否>50岁、病理类型和肿瘤直径均无关(P>0.05)。高表达LHPP组的卵巢癌细胞生长速度、克隆形成数、穿膜细胞数明显高于对照组。高表达LHPP后,上皮表型标志物表达下调,而间质表型标志物表达上调。结论:LHPP在上皮性卵巢癌中高表达,可能通过调控EMT过程,促进卵巢癌细胞的增殖、侵袭及迁移,有望成为上皮性卵巢癌的预后标志物及临床治疗靶点。Objective:To investigate effect and mechanism of phospholysine phosphohistidine inorganic pyrophosphate phosphatase(LHPP)on malignant behavior of epithelial ovarian cancer.Methods:The fresh tissue and paraffin sections of epithelial ovarian cancer and normal ovary in Qingdao Municipal Hospital from September 2016 to September 2019 were collected.The expression of LHPP in epithelial ovarian cancer and normal ovarian tissue was detected by immunohistochemical staining.The effect of LHPP on ovarian cancer cell proliferation was detected by CCK-8 and plate clonogenesis assay.Transwell test was used to detect the effect of LHPP on the invasion and migration of ovarian cancer cells.Western Blot assay was used to detect the expression of EMT related biomarkers.Results:The expression of LHPP in epithelial ovarian cancer was higher than that in normal ovarian tissue(P<0.01).The growth rate,numbers of colony formation and cells transfected into the subventricular layer of the high-expression LHPP group were significantly higher than that of the control group(P<0.05).Western Blot indicated that the expression of epithelial markers was down-regulated,while mesenchymal markers was up-regulated after LHPP over-expression.Conclusions:The high expression of LHPP in epithelial ovarian cancer promotes the proliferation,invasion and migration of ovarian cancer cells by regulating the process of epithelial mesenchymal transition,which is expected to be a prognostic marker and clinical treatment target of ovarian cancer.
