摘 要:目的:观察血管内皮生长因子(VEGF)和水通道蛋白4(AQP4)在脑膜瘤及其瘤周水肿中的表达,以探讨VEGF和AQP4在脑膜瘤瘤周水肿形成过程中的作用。方法:采集2012年至2018年于内蒙古自治区人民医院神经外科手术切除并病理证实的脑膜瘤40例,应用免疫组织化学法以及蛋白质印迹法(Western blot)检测脑膜瘤组织、瘤周组织和正常脑组织中VEGF及AQP4的表达,再通过反转录-聚合酶链反应(RT-PCR)法,检测VEGF的mRNA表达。采用t检验比较VEGF、AQP4及VEGF的mRNA表达强度在脑膜瘤组织、瘤周组织和正常脑组织中的差异,应用Pearson法进行相关性分析。结果:40例脑膜瘤组织中VEGF阳性表达率为91.3%;VEGF蛋白水平在脑膜瘤组织、瘤周水肿脑组织、正常脑组织中呈梯度降低关系(0.39±0.08、0.21±0.03、0.04±0.02)。在肿瘤组织中,VEGF的蛋白水平、RNA水平与瘤周水肿呈正相关( r=0.889、0.876, P值均<0.05);在瘤周水肿脑组织中,蛋白水平与瘤周水肿呈正相关( r=0.919, P<0.05)。AQP4在脑膜瘤及瘤周水肿组织中的表达均高于正常脑组织,而在瘤周水肿组织中的表达要高于在肿瘤组织中的表达,且两者与正常脑组织之间差异有统计学意义( χ2=40.102、46.875, P<0.05),而且AQP4的表达强度与瘤周水肿程度呈正相关。 结论:VEGF与AQP4共同参与瘤周水肿形成,可能存在协同作用。Objective To observe the expression of vascular endothelial growth factor(VEGF)and aquaporins 4(AQP4)in meningioma and peritumor brain edema(PTBE),and to explorethe the role of VEGF and APQ4 in the formation of PTBE.Methods The tissues of meningioma,a total of 40 patients,were collected from 2012 to 2018 in the department of neurosurgery of People’s Hospital of Inner Mongolia and pathology confirmed,application immune histochemical method and Western blotting method to detect meningioma tissues,PTBE tissues and nomal brain tissues in the protein expression of vascular endothelial growth factorand aquaporins 4,through the reverse transcription PCR method again,to detect the mRNA expression of vascular endothelial growth factor.Applied T test to compare the expression intensity of VEGF,AQP4 and mRNA of VEGF in meningioma tissues,PTBE tissues and normal brain tissues,and Pearson method was used for correlation analysis.Results VEGF in 40 cases with meningioma tissue positive expression rate is 91.3%;VEGF protein levels of tumor tissue,PTBE tissue and normal brain tissue are gradient lower relationship(0.39±0.08,0.21±0.03,0.04±0.02).In tumor tissue,VEGF levels of protein,RNA levels and PTBE were positively correlated(r=0.889,0.876,all P<0.05);In PTBE tissue,protein level was positively related with tumor weeks edema(r=0.919,P<0.05).The expression of AQP4 in meningioma and PTBE tissue were higher than in the normal brain tissue;and the expression of PTBE tissue is higher than expression in tumor tissue,and between the difference has statistically significant(χ2=40.102,46.875,all P<0.05),and the expression strength of AQP 4 were positively associated with the degree of edema.Conclusion VEGF and AQP4 jointly participate in the formation of peritumor brain edema,which may have a synergistic effect.
