摘 要:目的观察以叶酸为靶向分子担载紫杉醇纳米颗粒对肝癌细胞生物学行为的影响。方法常规培养肝癌细胞,将其分为对照组、空聚合物载体(PEG-b-PLA,简称为M)、紫杉醇(PTX)、高分子键合紫杉醇胶束(M(PTX))及叶酸靶向紫杉醇胶束(FA-M(PTX))。采用CCK-8法检测细胞活性,Annexin V-FITC/PI双染流式细胞术检测细胞凋亡。结果 PTX、M(PTX)和FA-M(PTX)这3种高分子键合胶束对肝癌细胞均有抑制作用,并且随着药物浓度、作用时间的增加,对细胞生长的抑制率明显增强(P<0.05)。结论以叶酸为靶向分子担载紫杉醇纳米颗粒具有较好的抗肝癌作用,可能是一种有潜力的紫杉醇缓控释新剂型用于提高其药效。Objective To observe effects of sequoia nanoparticles with folic acid on the biological behavior liver cancer cells.Methods Liver cells were divided into four groups,i.e,control,PEG-b-PLA(M),paclitaxel(PTX),polymer-bonded paclitaxel micelles(M(PTX))and sequoia nanoparticles with folic acid(FA-M(PTX)).CCK-8 assay was used to detect cell activity and apoptosis was observed by Annexin V-FITC/PI.Results CCK-8 showed that PTX,M(PTX)and FA-M(PTX)could inhibit the activity of liver cancer cells with the concentration and time.And also,Annexin V-FITC/PI double staining showed that all of them increase the apoptosis rate of liver cancer cells.Conclusion The acrylic-targeted sequoia nanoparticles have a better anti-liver cancer effect,and maybe a potential new agent of yew alcohol slow-control release to improve its efficacy.
