摘　　要：目的探讨长链非编码RNA (lncRNA) lnc-ZNF37A-2 (RP11-672F9.1,AL133271.1)对肺癌细胞迁移和侵袭能力的影响。方法利用GEPIA网站分析lnc-ZNF37A-2在癌症基因组图谱(TCGA)数据库不同分期肺癌组织中的表达,同时通过starBase v3.0分析lnc-ZNF37A-2表达量与肺癌患者总生存期的相关性。通过Co-LncRNA网站预测与lnc-ZNF37A-2相关的m RNA,进行基因富集和通路富集分析。干扰肺癌细胞中lnc-ZNF37A-2的表达,采用实时定量PCR检测lnc-ZNF37A-2的表达效率。采用划痕实验、Transwell侵袭实验检测细胞的迁移、侵袭能力。结果 lnc-ZNF37A-2在Ⅳ期肺癌患者中的表达高于Ⅰ~Ⅲ期患者,且lnc-ZNF37A-2高表达与总生存期呈负相关;lnc-ZNF37A-2相关的mRNA基因富集分析得到的通路与细胞黏附相关;沉默lnc-ZNF37A-2后肺癌细胞A549的迁移和侵袭能力明显降低。结论 lnc-ZNF37A-2在肺癌细胞中起到癌基因的作用,与lnc-ZNF37A-2共表达的m RNA影响了肿瘤微环境中细胞黏附和细胞外基质受体的相互作用,沉默lnc-ZNF37A-2抑制了肺癌细胞迁移和侵袭能力。Objective To investigate the effects of long non-coding RNA(lncRNA)lnc-ZNF37A-2(RP11-672F9.1,AL133271.1)on the invasion and migration abilities of lung cancer cells.Methods GEPIA website was used to analyze lnc-ZNF37A-2 expression at different pathological stages in lung cancer samples,using the data acquired from The Cancer Genome Atlas(TCGA)database.The relationship between lnc-ZNF37A-2 expression and overall survival(OS)of lung cancer patients was assessed using the starBase v3.0 program.Geneset and pathway enrichment analyses were conducted to evaluate the potential functions of mRNAs co-expressed with lnc-ZNF37A-2 that were predicted using the Co-LncRNA program.We used real-time quantitative PCR to assess silencing efficiency after performing lnc-ZNF37A-2 silencing in the lung cancer cells.Wound healing and Transwell invasion assays were performed to assess migration and invasion abilities of the cancer cells.Results lnc-ZNF37A-2 expression was significantly increased in stageⅣlung cancer patients than in stageⅠtoⅢlung cancer patients.Survival analysis revealed that increased lnc-ZNF37A-2 expression might be related to decreased OS in lung cancer patients.Gene enrichment analysis of lnc-ZNF37A-2-related mRNAs indicated that lnc-ZNF37A-2 majorly affects genes involved in cell adhesion.Moreover,silencing of lnc-ZNF37A-2 reduced the migration and invasion abilities of lung cancer cells.Conclusion lnc-ZNF37A-2 plays a crucial role in regulating the expression of oncogenes in lung cancer cells.The mRNAs co-expressed with lnc-ZNF37A-2 affect cell adhesion and extracellular matrix receptor interaction in tumor microenvironment.Furthermore,silencing of lnc-ZNF37A-2 leads to the inhibition of migration and invasion of lung cancer cells.
