摘  要:目的:探索中性粒细胞来源外泌体(PMN-Exo)体外诱导人骨髓间充质干细胞(hBMSCs)分泌促血管生成的相关因子。方法:免疫磁珠法提取并超离法分离PMN-Exo,对PMN-Exo进行鉴定及内吞实验;细胞实验分为PBS组和PMN-Exo组,EdU法检测细胞增殖活性,划痕实验检测细胞迁移能力,并对PMN-Exo诱导hBMSCs分泌成血管相关因子的能力进行检测。结果:成功分离鉴定PMN-Exo,并证实PMN可有效进入hBMSCs。PMN-Exo组与PBS组比较,细胞增殖活性、迁移能力明显增强,并大量合成VEGF和PDGF。结论:PMN-Exo可显著促进hBMSCs的增殖和迁移能力,促进其成血管相关因子的表达。Objective: To study the effects of PMN-derived exosomes(PMN-Exo) on the promotion of angiogenesis-related factors of hBMSCs in vitro. Methods: PMNs from the mouse marrow were extracted by magnetic activated cell sorting. PMN-Exo obtained by supercentrifugation and PMN-Exo internalization into hBMSCs were identified. hBMSCs were treated with PMN-Exo(PMN-Exo group) and PBS(PBS group) respectively. The promotion effect of PMN-Exo on the expression of VEGF and PDGF of hBMSCs was studied. Cell proliferation of hBMSCs were detected by EdU staining. Cell migration was assessed by scratch test. The mRNA expression of VEGF and PDGF of hBMSCs was analyzed by QPCR and WB. Results: PMN-Exo was isolated and conformed by the identification. Compared with PBS group, PMN-Exo group showed stronger proliferation, cell migration and higher expression of VEGF and PDGF. Conclusion: PMN-Exo can promote the proliferation, migration and the expression of angiogenesis-related factors of hBMSCs.
