摘 要:目的观察结膜下注射色素上皮衍生因子基因修饰的人脐带间充质干细胞(PEDF-MSCs)对糖尿病大鼠角膜上皮损伤的修复作用及其对上皮下神经表达改变的影响。方法取32只健康SD雄性大鼠,将其随机分为正常对照组和实验组。实验组建造2型糖尿病大鼠模型,分为磷酸盐缓冲液组(PBS)、hUCMSCs治疗组、PEDF-MSCs治疗组。建造角膜上皮损伤模型,并进行球结膜下注射相应溶液干预治疗。观察角膜上皮缺损修复面积、泪液分泌量和角膜上皮下神经密度。结果①糖尿病组大鼠较正常对照组角膜上皮修复延迟,泪液分泌量少,角膜上皮下神经β-tubulinⅢ的表达低,上皮下神经密度低。②经PEDF-MSCs,hUCMSCs治疗组与实验对照组相比,角膜荧光素钠染色面积明显减少,大鼠泪液分泌量增多,β-tubulinⅢ的表达高,上皮下神经密度高,且PEDF-MSCs治疗组效果强于hUCMSCs治疗组,各组两两比较,差异均有显著性(P<0.001)。结论结膜下注射PEDF-MSCs可促进糖尿病大鼠角膜损伤修复及角膜上皮下神经表达,保护眼表组织。Objective To observe the effect of subconjunctival injection of human umbilical cord-derived mesenchymal stem cells modified by pigment epithelial-derived factor gene on corneal epithelial injury and the expression of corneal subbasal nerve in diabetic rats,and to explore the protective effect of PEDF-MSCs on corneal Keratopathy.Methods Thirty two healthy male SD rats were randomly divided into normal control group and experimental group.The experimental group established type 2 diabete model and was divided into PBS group,hUCMSCs treatment group and PEDF MSCs treatment group.The model of corneal epithelial injury was established and the corresponding solution was injected under the bulbar conjunctiva.The repair area of corneal epithelial defect,the amount of lacrimal secretion and the density of the superior subcutaneous nerve were observed.Results①Compared with the normal control group,the diabetic rats had delayed corneal epithelial repair,less secretion of tears,lower expression of corneal epithelial nerveβ-tubulinⅢ,and lower density of subepithelial nerve.②PEDF MSCs group,hUCMSCs group compared with the experimental control group,the area of corneal fluorescein sodium staining was significantly reduced,the lacrimal secretion was increased,the expression ofβ-tubulinⅢand the density of subepithelial nerve were high,and the effect of PEDF MSCs treatment group was better than that of hUCMSCs treatment group,which has significant difference(P<0.001).Conclusion The subconjunctival injection of PEDF MSCs can accelerate the repair of corneal injury and the expression of subcutaneous nerve in the cornea of diabetic rats,and protect the ocular surface tissue.
