摘 要:目的探讨安全剂量范围内不同剂量亚甲基蓝(MB)对视网膜缺血再灌注损伤(RIRI)大鼠视网膜细胞凋亡的影响。方法取健康成年S-D大鼠,随机分为正常对照组、生理盐水组与MB组,MB组按用药剂量不同,分为1mg/kg MB,2mg/kg MB,3mg/kg MB,4mg/kg MB 4个亚组。采用高眼压法建立RIRI大鼠模型,于造模前连续3d及造模前30min在MB各亚组大鼠腹腔内分别注射1mg/kg MB,2mg/kg MB,3mg/kg MB,4mg/kg MB,生理盐水组腹腔注射同等剂量生理盐水,于造模成功后24h处死大鼠,免疫组织化学检测各组Bcl-2,Bax阳性细胞表达情况。结果正常对照组大鼠视网膜可见大量Bcl-2阳性细胞,3mg/kg MB组较生理盐水组及其他浓度组大鼠视网膜Bcl-2阳性细胞数明显增多,差异均有显著性(均P<0.05)。正常对照组大鼠视网膜几乎未见Bax阳性细胞,在各实验组中,3mg/kg MB组及4mg/kg MB组大鼠视网膜Bax阳性细胞数目最少,两组阳性细胞数目变化不显著,差异均有显著性(均P<0.05)。结论MB可减轻RIRI大鼠视网膜细胞凋亡,具有神经保护作用,其中以浓度为3mg/kg MB最佳。Objective To explore the effects of different doses of methylene blue on retinal apoptosis in rats with retinal ischemia-reperfusion injury in a safe dose range.Methods Health adult S-D rats were randomly divided into normal control group,Normal Saline group and MB group.The MB group was divided into 1 mg/kg MB,2 mg/kg MB,3 mg/kg MB,4 mg/kg MB sub-groups according to different dosages.The RIRI rat model was made by elevating the intraocular pressure method.The RIRI rat model was established by the method high intraocular pressure method,and the intraperitoneal injection was performed in each(1 mg/kg,2 mg/kg,3 mg/kg,4 mg/kg in each subgroup)3 days before and 30 minutes before the models were established.The same dose of NS was injected intraperitoneally in NS group.The rats were sacrificed 24 h after the models were successfully developed,and the expression of Bcl-2 and Bax positive cells in each group was detected by immunohistochemical detection.Results In the normal control group,there are a larg number of Bcl-2 positive cells.3 mg/kg MB group compared with strok-physiological saline solution group and other concentration,rat retina Bcl-2 positive cells was significantly increased.The differences were statistically significant(all P<0.05).The content of Bax positive cells in the normal control group were rare.In the experimental group,the rat retina Bax positive cells in 3 mg/kg MB group and 4 mg/kg MB group were the least,two groups of positive cell number change was not significant.The differences were statistically significant(all P<0.05).Conclusion MB can reduce the degree of retinal cell apoptosis in rats with RIRI,which has the nerve protective effect,and 3 mg/kg MB is the best therapeutic regimen.
