摘 要:目的研究2,4,6-三硝基苯磺酸(TNBS)诱导灌肠法建立大鼠炎症性肠病模型的可靠性及稳定性。方法将40只SD大鼠随机分为2组,TNBS组20只大鼠给予TNBS-50%乙醇混合溶液灌肠1次,乙醇对照组20只大鼠给予等体积50%乙醇溶液灌肠1次,观察2组大鼠体质量变化、存活率、结肠大体形态及结肠组织病理学改变,确立IBD模型是否建立成功。结果TNBS组建模第3天体质量下降达到最大程度,随着时间的推移,大鼠体质量缓慢恢复,而乙醇对照组大鼠的体质量一直呈稳定增长的状态;建模7d内,TNBS组大鼠存活率为70%(14/20),至建模14 d无死亡,乙醇对照组大鼠建模14 d内均无死亡。造模第7天,TNBS组结肠上皮细胞出现坏死脱落,肌层出现坏死;乙醇对照组未见明显变化。造模第14天,TNBS组大鼠有结肠变短的现象,且结肠组织结构明显破坏;乙醇对照组大鼠结肠结构没有破坏及病变迹象。TNBS组造模第7天和第14天结肠大体评分和病理组织学评分均明显高于乙醇对照组(P均<0.05),且造模第7天评分均明显高于第14天(P均<0.05)。结论TNBS联合乙醇诱导灌肠法建立的炎症性肠病模型稳定,在造模后的第7天炎症反应最明显,适于炎症性肠病慢性炎症反应的实验室研究。Objective It is to study the reliability and stability of 2,4,6-trinitrobenzenesulfonic acid(TNBS)induction enema to establish a rat model of inflammatory bowel disease.Methods 40 SD rats were randomly divided into 2 groups.20 rats in the TNBS group were given TNBS-50%ethanol mixed solution once,and 20 rats in the ethanol control group were given an equal volume of 50%ethanol solution once.The body mass changes,survival rate,gross morphology of the colon and histopathological changes of the colon in the two groups of rats were observed to confirm whether the IBD model was established successfully or not.Results The body weight of the TNBS group decreased to the maximum on the third day of modeling.As time passed,the body weight of the rats recovered slowly,while the body weight of the ethanol control group was increasing steadily;within 7 days of modeling,the survival rate of the rats in the TNBS group was 70%(14/20),and there was no death within 14 days of modeling,and there was no death in the ethanol control group within 14 days of modeling.On the 7th day of modeling,the colonic epithelial cells in the TNBS group appeared necrosis and shedding,and the muscle layer appeared necrosis;the colon structure of the rats in the ethanol control group had no damage or signs of disease.On the 14th day of modeling,the rats in the TNBS group had their colon shortening,and the structure of the colon tissue was obviously damaged;the colon structure of the rats in the ethanol control group had no damage or signs of disease.The gross and histopathological scores of the colon in the TNBS group were significantly higher than those of the ethanol control group on the 7th and 14th day of the model building(P<0.05),and the scores on the 7th day of the model building were significantly higher than those on the 14th day(P both<0.05).Conclusion The inflammatory bowel disease model established by TNBS combined with ethanol-induced enema is stable,and the inflammatory response is most obvious on the 7th day after modeling,which is suita
