摘 要:目的探讨分子生物学技术在区分ABO亚型与白血病致ABO血型抗原减弱中的应用。方法对1例表现为ABO血型抗原减弱的白血病患者,分别采用实时荧光定量PCR、序列特异性引物引导的PCR法(PCR-sequence specific primer,PCR-SSP)以及基因测序技术鉴定其ABO血型。结果实时荧光定量PCR以及PCR-SSP的检测结果均表明该患者ABO血型为A2B,测序结果确定其ABO基因型为A205/B101,该患者为ABO亚型且存在因白血病致血型抗原减弱。结论利用分子生物学技术能在基因水平上准确鉴定ABO血型,对保障临床安全输血具有重要意义。Objective To explore the application of molecular biology techniques in identifying the ABO blood subtype and the ABO antigen reducing in leukemia patients.Methods For a case of weak ABO antigen patient with leukemia,the ABO blood group was detected by real-time fluorescence quantitative PCR(RT-PCR),PCR-sequence specific primer(PCR-SSP)and gene sequencing,respectively.Results RT-PCR and PCR-SSP results showed that the patient’s ABO blood group was A2 B and the genotype was A205/B101 by further sequencing analysis.These results indicated that the patient was ABO subgroup with ABO antigen reducing affected by leukemia.Conclusions ABO blood group can be accurately identified at the genetic level by molecular biology methods,which has important significance to guarantee clinical transfusion safety.
