摘 要:目的从通路水平探讨乳腺癌发病的相关基因。方法使用i-GSEA4GWASv2和ICSNPathway对GRASPv2.0中乳腺癌的GWAS数据进行基因富集分析,有意义的GO通路依照GO数据库注释构建有向无环图。结果与乳腺癌发生有关的4条通路:小GTP酶介导的信号转导(P平均=0.002;FDR平均=0.017)、GnRH信号通路(P_(平均)=0.005;FDR_(平均)=0.041)、MAPK信号通路(P_(平均)=0.001;FDR_(平均)=0.026)、长时程增强通路(P_(平均)=0.002;FDR_(平均)=0.025)。结论为乳腺癌发病机制从小GTP酶信号转导、刺激信号传递、性腺激素释放激素方面提供了新的思路。Objective To explore the genes related to breast cancer at the pathway level.Methods Gene enrichment analysis of breast cancer GWAS data in GRASPv2.0 was performed using i-GSEA4GWASv2 and ICSNPathway.Directed acyclic graphs were constructed according to GO database annotations.Results Four pathways related to breast cancer:small GTPase-mediated signal transduction(Paverage=0.002;FDR average=0.017),GnRH signal pathway(Paverage=0.005;FDR average=0.041),MAPK signal pathway(Paverage=0.001;FDRaverage=0.026),long-term enhancement pathway(Paverage=0.002;FDRaverage=0.025).Conclusion It provides new ideas for breast cancer pathogenesis from small GTPase signal transduction,stimulation signal transmission,and gonadotropin releasing hormone.
