摘 要:目的探索花椒籽油对大鼠烧伤模型创面愈合及血清炎症因子调节的作用。方法用铜梳烧伤法构建大鼠烧伤模型。将烧伤模型大鼠随机分为低、高剂量实验组和对照组,假手术组将梳子在37℃温水中浸入3 min后进行烧伤处理,每组8只。低、高剂量实验组分别于烧伤处涂抹花椒籽油0.5和1.0 mL;对照组和假手术组均给予烧伤处涂抹0.9%NaCl 1.0 mL。4组大鼠每天给药1次,均连续干预21 d。用荧光分析法检测烧伤皮肤组织中硫代巴比妥酸反应物(TBARS)和还原型谷胱甘肽(GSH)的水平,用酶联免疫吸附法检测血清炎症因子的水平。结果干预后21 d,假手术组、对照组和低、高剂量实验组的TBARS分别为(0.26±0.04),(1.69±0.21),(0.96±0.04)和(0.56±0.08)mmol·mL^-1,GSH分别为(1.15±0.21),(0.28±0.06),(0.58±0.06)和(0.78±0.05)mmol·mL^-1,白细胞介素-6分别为(10.75±0.02),(1523.25±253.08),(698.25±12.36)和(452.16±36.28)ng·L^-1,肿瘤坏死因子-α分别为(8.35±0.04),(1756.35±234.28),(762.68±34.69)和(523.16±34.05)ng·L^-1。对照组的上述指标与假手术组和低、高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论花椒籽油可以通过降低炎性因子水平而促进大鼠烧伤创面的愈合。Objective To explore the effects of Zanthoxylum bungeanum seed oil on wound healing and serum inflammatory factors in rats with burns.Methods A copper comb burn method was used to construct a rat burn model.The burn model rats were randomly divided into experimental-L/-H groups and control group.Sham-operation group was immersed in comb at 37℃for 3 minutes.There were 8 rats in each group.Experimental-L/-H groups were treated with 0.5 and 1.0 mL of pepper seed oil at the burns,respectively.Control and sham-operation groups were treated with 0.9%NaCl 1.0 mL at the burns.The rats in the four groups were administered once a day for 21 days.The levels of thiobarbituric acid reactant(TBARS)and reduced glutathione(GSH)in the burn skin tissue were detected by fluorescence analysis,and the serum inflammatory factors were detected by enzyme-linked immunosorbent assay.Results After intervention 21 days,the TBARS levels in sham-operation group,control group and experimental-L-H groups were(0.26±0.04),(1.69±0.21),(0.96±0.04)and(0.56±0.08)mmol·mL^-1,GSH were(1.15±0.21),(0.28±0.06),(0.58±0.06)and(0.78±0.05)mmol·mL^-1,interleukin-6 were(10.75±0.02),(1523.25±253.08),(698.25±12.36)and(452.16±36.28)ng·L^-1,tumor necrosis factor-αwere(8.35±0.04),(1756.35±234.28),(762.68±34.69)and(523.16±34.05)ng·L^-1.The above-mentioned indexes in the control group were significantly different from those in the sham-operation group and experimental-L,-H groups(all P<0.05).Conclusion Zanthoxylum bungeanum seed oil can promote the wound healing of burn in rats by reducing the level of inflammatory factors.
