摘 要:目的探讨少突胶质细胞转录因子2 (oligodendrocyte transcription factor 2,Olig2)在胶质母细胞瘤(glioblastoma,GBM)的表达意义。方法采用组织微阵列免疫组化法检测277例GBM组织标本(每例标本选择5个取材区域)及12例正常脑组织中Olig2表达情况。比较每例标本不同取材区域的Olig2表达差异,分析Olig2表达情况与临床病理参数间的相关性。结果 Olig2在正常脑组织与GBM的阳性信号均位于细胞核。GBM Olig2阳性细胞比例为60.0%(Q25-Q75:10.0%~80.0%)。Olig2表达与病人年龄、GBM病理类型、IDH1R132H突变及PDGFRα表达显著相关(均P <0.05)。在Olig2阳性表达的GBM中,34.7%肿瘤组织内显示明显异质性。Olig2表达与预后明显正相关(P=0.030)。联合TERT启动子区突变及IDH1R132H突变可更好对GBM进行分子分型。结论 Olig2在GBM表达差异性较大,高表达者预后较好。检测Olig2有助于判断GBM异质性及肿瘤细胞起源,可能具有指导临床诊疗的价值。Objective To explore the significance of expression of Olig2 protein in glioblastoma(GBM). Methods The tissue microarray immunohistochemistry was used to detect the expression of Olig2 in 277 GBM tissue specimens(5 selected regions for each specimen)and 12 normal brain tissues. The differences in Olig2 expression were compared between the different sample areas of each specimen,and the correlation between Olig2 expression and clinicopathological parameters, and prognosis of GBM were analyzed. Results The positive signals of Olig2 in normal brain tissue and GBM were located in the nucleus. The proportion of Olig2 positive cells in GBM was 60.0%(Q25-Q75: 10.0%-80.0%). The expression of Olig2 was significantly correlated with patient age, GBM pathological type,IDH1 R132 Hmutation and PDGFRα expression(all P <0.05). Among the GBM with positive expression of Olig2, 34.7% of tumor tissues showed obvious heterogeneity. The significant positive correlation existed between Olig2 expression and prognosis( P = 0.030).Combining TERT promoter mutation and IDH1 R132 Hmutation could perform better molecular typing of GBM. Conclusions The expression of Olig2 in GBM is different, and those with high expression have a better prognosis. The detection of Olig2 helps to determine the heterogeneity of GBM and origin of tumor cells, and may have the value of guiding clinical diagnosis and treatment.
