摘 要:本试验旨在建立缺硒大鼠模型,研究长期缺硒对大鼠体内D-丝氨酸、L-丝氨酸水平的影响。选取3周龄雄性断乳大鼠12只,随机分为2组(每组6只):缺硒组大鼠饲喂低硒饲粮(硒水平为0.02 mg/kg),对照组大鼠同期饲喂正常饲粮(硒水平为0.18 mg/kg),试验期300 d。利用电感耦合等离子体质谱法测定血浆中硒水平,利用邻苯二甲醛和N-乙酰-L-半胱氨酸柱前衍生,高效液相色谱法测定大鼠体内D-丝氨酸、L-丝氨酸水平。结果表明:1)使用Agilent ZORBAX SB-C18色谱柱(4.6 mm×150 mm,5μm),采用50 mmol/L醋酸铵和甲醇作为流动相进行梯度洗脱,D-丝氨酸和L-丝氨酸得到很好地分离。2)与饲喂正常饲粮的大鼠相比,饲喂低硒饲粮的大鼠血浆中硒水平极显著降低(P<0.01),血浆中D-丝氨酸、L-丝氨酸以及小脑中L-丝氨酸水平升高,但差异未达显著水平(P>0.05)。由此可见,通过饲喂低硒饲粮可建立缺硒大鼠模型,长期低硒饲粮饲喂对大鼠血浆、小脑中D-丝氨酸、L-丝氨酸水平无显著影响。This experiment was conducted to establish a selenium deficiency rat model and investigate effects of long-term selenium deficiency on D-serine and L-serine levels in rats.Twelve male weaned rats aged 3 weeks were randomly divided into 2 groups and each group had 6 rats.Rats in the selenium deficiency group were fed a low selenium diet(selenium level of 0.02 mg/kg)and rats in the control group were fed a normal diet(selenium level of 0.18 mg/kg).The experiment lasted for 300 days.Selenium level in plasma were determined using inductively coupled plasma mass spectrometry.High performance liquid chromatography was used to analyze the levels of D-serine and L-serine after derivatizating with o-phthalaldehyde and N-acetyl-L-cysteine.The results showed as follows:1)under the mobile phase consisting of 50 mmol/L of ammonium acetate and methanol,D-serine and L-serine were successfully separated on the Agilent ZORBAX SB-C18 column(4.6 mm×150 mm,5μm).2)Compared with the rats fed normal diet,plasma selenium level of rats fed selenium deficiency diet was significantly decreased(P<0.01),and plasma D-serine,L-serine levels as well as cerebellum L-serine level were increased,but differences were not significant(P>0.05).In conclusion,the selenium deficiency rat model is successfully established by selenium-deficient diets,and D-serine,L-serine levels in plasma and cerebellum are not significantly affected by the long-term selenium deficiency.
