摘 要:目的探讨瓜氨酸(Cit)对溃疡性结肠炎(UC)大鼠的保护作用及对外周血单核细胞的影响。方法将15只Wistar雄性大鼠随机分为正常组、UC模型组和Cit给药组,采用2,4,6-三硝基苯磺酸(TNBS)/乙醇法进行造模,造模成功后连续灌胃给药7 d,观察三组大鼠体质量变化、疾病活动指数(DAI),取结肠组织进行病理学观察,取外周血进行单核细胞计数及比例检测。结果 UC模型组大鼠造模后7 d体质量显著降低(P<0.05),而Cit给药组大鼠体质量变化差异无统计学意义(P>0.05)。Cit给药组大鼠DAI评分较UC模型组低,但两组之间差异无统计学意义(P>0.05)。结肠组织HE染色结果显示,补充Cit可以减轻UC大鼠结肠肠管的炎症损伤程度。补充Cit的UC大鼠血液中单核细胞计数和单核细胞比例明显低于UC模型组大鼠。结论补充Cit对UC肠道的炎症损伤具有保护作用,Cit可以降低UC大鼠外周血单核细胞的数量及比例。Objective To investigate the protective effect of citrulline(Cit) on ulcerative colitis(UC) rats and the influence of peripheral blood mononuclear cells. Methods Fifteen Wistar male rats were randomly divided into normal group, UC model group and UC+Cit group. 2,4,6-trinitrobenzenesulfonic acid(TNBS)/ethanol method was used to establish UC rat model. After continuous gastric administration for 7 days, rat body weight changes and disease activity index(DAI) were observed. Rat colon tissue was collected for pathological observation. Rat peripheral blood was collected for monocyte counting and proportional detection. Results The weight of rats in UC model group was significantly decreased 7 days after model establishment(P<0.05), but there was no significant difference in body weight of the UC+Cit group(P>0.05). The DAI score of the UC+Cit group was lower than that of the UC model group, but there was no statistical difference between the two groups(P>0.05). HE staining of colon tissue showed that supplementation of Cit could reduce the degree of inflammatory damage in the colon of UC rats. In addition, the count of monocytes as well as the proportion of monocytes in the blood of UC+ Cit group rats was significantly lower than that of UC model group. Conclusion Supplementation of Cit has protective effect on intestinal inflammatory injury of UC. Cit supplementation can reduce the number and proportion of monocytes in peripheral blood of UC rats.
