摘 要:为了全面获得和发掘薏苡的基因数据、功能信息以及潜在的SSR标记,本研究采用PacBio SequelTM第三代测序平台对干旱胁迫下薏苡苗期叶片进行全长转录组测序与分析,获得总计98 858条非冗余的高质量Unigenes,其平均长度为2 318 bp,1 kb以上Unigenes占91.74%。利用BLAST等软件在NR、Swiss Prot、eggNOG、GO和KEGG等公共数据中注释了90 339个Unigenes,占91.50%。NR注释表明,在90 093个被注释的Unigenes中,与之匹配数量最多是来源于高粱的序列,其次是玉米和谷子,这与它们之间在进化上的亲缘关系相一致。GO注释显示,34 765个Unigenes被注释到生物过程、细胞组分和分子功能等三大类别;eggNOG注释的89 158条Unigenes分属于25个功能聚类;KEGG代谢通路分析表明,30 579个Unigenes被注释到133条已知代谢通路,其中,1 884条Unigenes涉及信号转导和环境适应途径,包括植物激素信号转导途径、MAPK信号途径和植物-病原互作信号途径,上述代谢通路或途径均与植物响应干旱等逆境胁迫相关。此外,利用MISA程序在38 878个Unigenes中检测到56 540个SSR位点。研究结果将为薏苡耐旱等抗逆基因发掘与克隆、抗逆分子生理机制解析以及分子标记开发提供科学依据和理论参考。In order to obtain the genetic data,functional information and potential SSR markers in Coix lachrymajobi,the full-length transcriptome sequencing and functional annotation analysis of Coix young leaves under drought stress were performed using Pac Bio SequelTMsequencing platform.In total,98 858 Unigenes were obtained with an average length of 2 318 bp,and the Unigenes with the length over 1 kb accounted for 91.74%.The Unigenes were further annotated into the public databases of NR,SwissProt,eggNOG,GO and KEGG.NR annotation revealed 90 093 Unigenes,and the number of related genes annotated with Sorghum bicolor was the highest,followed by Zea mays and Setaria italica,which were consistent with their evolutionary relationships.GO analysis showed that 34 765 Unigenes were annotated into biological process,cellular components and molecular function.eggNOG analysis revealed that 89 158 Unigenes were annotated into 25 functional clustering.KEGG analysis found that 30 579 Unigenes were annotated into 133 metabolic pathways.Among them,the total 1 884 Unig enes were enriched in signal transduction and environmental adaption pathways,including plant hormone signal transduction,MAPK signaling pathway and plant-pathogen interaction,which were related to plant responses to stress.In addition,a total of 56 540 SSR loci were found in 38 878 Unigenes by using the MISA program.This study will provide the foundation and reference for functional genome studies in Coix,especially on cloning of drought tolerance related-genes,clarifying their molecular and physiological mechanisms on stress tolerance,as well as developing SSR molecular markers.
