长链非编码RNA H19基因多态性与胃癌遗传易感性关系的研究

摘　　要：目的探讨长链非编码RNA H19(lncRNA H19)基因多态性与胃癌易感性的关系。方法采用聚合酶链反应(PCR)-RFLP技术检测128例胃癌患者与130例健康人群lncRNA H19 rs217727 C﹥T基因多态性的分布频率,通过不同基因模型分析其与胃癌发生及临床特征的关系。结果lncRNA H19 rs217727 C﹥T在对照组与病例组中进行不同基因模型下的Logistic回归分析结果显示,TT基因型增加胃癌的发病风险,OR=2.76,95%CI=1.28~5.94,P=0.009,校正混杂因素后OR=2.99,95%CI=1.37~6.56,P=0.006;等位基因模型中,T等位基因显著增加胃癌的发病风险,OR=1.65,95%CI=1.16~2.36,P=0.006。lncRNA H19 rs217727与胃癌分化程度、临床分期、淋巴结转移、远处转移均无关(P﹥0.05)。结论在广安汉族人群中lncRNA H19 rs217727 C﹥T多态位点与人群胃癌遗传易感性有关,TT基因型、T等位基因增加广安地区人群患胃癌的风险。Objective To investigate the association of long non-coding RNA H19(lncRNA H19)polymorphism with susceptibility to gastric cancer.Method The distribution frequency of lncRNA H19 rs217727 C>T gene polymorphism was detected by polymerase chain reaction(PCR)-RFLP in 128 patients with gastric cancer and 130 healthy people,and the association with the occurrence and clinical characteristics of gastric cancer was analyzed by different gene models.Result Logistic regression analysis of lncRNA H19 rs217727 C>T in different gene models showed that TT genotype(OR=2.76,95%CI=1.28-5.94,P=0.009,adjusted OR=2.99,95%CI=1.37-6.56,P=0.006)and T allele(OR=1.65,95%CI=1.16-2.36,P=0.006)were significantly associated with the increased risk of gastric cancer.There was no significant association of lncRNA H19 rs217727 with differentiation of gastric cancer,clinical stage,lymph node metastasis and distant metastasis(P>0.05).Conclusion The polymorphism of lncRNA H19 rs217727 C>T is associated with genetic susceptibility of gastric cancer in Guang’an Han population.TT genotype and T allele increase the risk of gastric cancer in Guang’an population.
