摘　　要：目的探讨miRNA-342表达与膀胱癌患者预后的关系。方法收集200例原发性膀胱癌患者的术后膀胱癌组织和邻近正常膀胱组织,采用实时聚合酶链反应(RT-PCR)检测膀胱癌组织和正常膀胱组织中miRNA-342的表达,收集患者临床资料,定期随访,分析miRNA-342表达与患者临床特征的关系,并分析不同miRNA-342表达水平的膀胱癌患者的生存情况,采用Cox回归分析评价膀胱癌患者预后的影响因素。结果膀胱癌组织中miRNA-342相对表达量为(3.42±0.61),明显低于正常膀胱组织中的(7.31±1.21),差异有统计学意义(P﹤0.01)。不同肿瘤分期及是否淋巴结转移和复发膀胱癌患者的膀胱癌组织中miRNA-342表达情况比较,差异均有统计学意义(P﹤0.05)。miRNA-342高表达患者的生存情况明显优于miRNA-342低表达患者,差异有统计学意义(P﹤0.01)。多因素分析表明,肿瘤分期、淋巴结转移、复发、miRNA-342表达是膀胱癌患者预后的独立影响因素(P﹤0.05)。结论miRNA-342有望成为新的膀胱癌预后生物标志物。Objective To investigate the relationship between miRNA-342 expression and prognosis in patients with bladder cancer.Method The bladder cancer tissue and adjacent normal tissue specimens from 200 patients with primary bladder cancer were collected,and the miRNA-342 expression in these specimens were detected by real-time polymerase chain reaction(RT-PCR),patients’clinical data were collected,and regular follow-up was administered for these patients,to analyze the relationship between miRNA-342 expression and patients’clinical characteristics,the survival of bladder cancer patients with varying miRNA-342 expression was investigated,and Cox regression analysis was used to evaluate the factors influencing the prognosis of bladder cancer patients.Result The relative expression of miRNA-342 in bladder cancer tissues was(3.42±0.61),which was significantly lower than that in normal bladder tissues at(7.31±1.21)(P<0.01).The expression of miRNA-342 in bladder cancer tissues was correlated with tumor stage,lymph node metastasis and recurrence(P<0.05).Better survival was noted among patients with high miRNA-342 expression compared to those with low miRNA-342 expression(P<0.01).Multivariate analysis showed that,tumor stage,lymph node metastasis,recurrence,and miRNA-342 expression were independent prognostic factors for the prognosis of bladder cancer patients(P<0.05).Conclusion miRNA-342 is expected to be a new prognostic biomarker for bladder cancer.
