摘 要:目的探讨甘草甜素对周围神经损伤模型大鼠的干预效果及其可能机制。方法选取50只SD健康雄性大鼠,建立周围神经损伤模型,随机分为对照组、模型组、低剂量、中剂量、高剂量组各10只。低、中、高剂量组大鼠分别给予10 mg/kg、20 mg/kg、30 mg/kg甘草甜素溶液灌胃,对照组、模型组大鼠灌胃等体积生理盐水。检测各组大鼠有髓神经纤维直径和数量,计算比目鱼肌肌肉指数;并检测脊髓组织生长相关蛋白43(GAP-43)、神经营养因子受体p75(p75NTR)蛋白表达水平,T、B淋巴细胞增殖率,神经元细胞凋亡率。结果模型组、低剂量组、中剂量组、高剂量组、对照组大鼠脊髓GAP-43相对表达量依次升高,p75NTR相对表达量依次降低,比目鱼肌肌肉指数及有髓神经纤维直径、数量依次升高(P<0.05);各个时间段,模型组、低剂量组、中剂量组、高剂量组、对照组大鼠T、B淋巴细胞增殖率、神经元细胞凋亡率依次降低(P<0.05)。结论甘草甜素可能通过调控GAP-43、p75NTR蛋白的表达,来抑制周围神经损伤模型大鼠T、B淋巴细胞增殖以及神经元细胞凋亡,从而促进周围神经再生,减轻比目鱼肌萎缩程度。Objective To explore the intervention effect of glycyrrhizin on nerve regeneration in rat model of peripheral nerve injury and its potential mechanism.Methods Fifty SD healthy male rats were enrolled to establish the model of peripheral nerve injury,and the rats were randomly divided into control group,model group,low dose group,moderate dose group and high dose group,with 10 rats in each group.Rats in the low,middle and high dose groups were given 10 mg/kg,20 mg/kg and 30 mg/kg glycyrrhizin solution by gavage,respectively,and rats in the control group and the model group were gavaged with isovolumetric normal saline.The diameter and number of myelinated nerve fibers of rats were determined in each group,muscle index of soleus muscle was calculated;in addition,the expression levels of spinal tissue growth-associated protein 43(GAP-43)and p75 neurotrophin receptor(p75NTR)protein,the proliferation rates of T and B lymphocytes,and the apoptosis rate of neurons were detected.Results In the model group,the low dose group,the moderate dose group,the high dose group and the control group,the relative expression of spinal GAP-43 increased in turn,the relative expression of p75NTR decreased in turn,and muscle index of soleus muscle as well as the diameter and number of myelinated nerve fibers increased in turn(P<0.05).At each time interval,the proliferation rate of T or B lymphocytes as well as the apoptosis rate of neurons decreased successively in the model group,the low dose group,the moderate dose group,the high dose group and the control group(P<0.05).Conclusion Glycyrrhizin may inhibit the proliferation of T and B lymphocytes and neuronal cell apoptosis in rat model of peripheral nerve injury by regulating the expression of GAP-43 and p75NTR proteins,thus promoting peripheral nerve regeneration and reducing the degree of soleus muscle atrophy.
